摘 要:目的探讨外源性核苷酸对乙醇发育毒性的干预作用。方法采用啮齿类动物植入后全胚胎培养技术,将胎龄8.5 d的ICR小鼠胚胎以4.0g/L乙醇染毒,然后用不同剂量(0.16mg/L,4mg/L,100mg/L)的外源性核苷酸干预。培养48 h后对小鼠胚胎生长发育和组织器官形态分化进行评分。结果剂量为0.16mg/L及100mg/L的核苷酸干预可使胚胎颅臀长增大,体位、后神经管评分明显升高,卵黄囊直径增大,且与阴性对照组相比差异无统计学意义。中剂量(4mg/L)核苷酸干预还可使头长增大,对体位、心脏、后神经管、后脑、中脑、前脑、听觉、视觉、嗅觉、前肢芽及后肢芽均有改善,且与阴性对照组相比差异无统计学意义。结论外源性核苷酸可能有拮抗乙醇发育毒性的作用。OBJECTIVE To evaluate the effect of nucleotide supplementation on ethanol-induced embryonic abnormality in vitro.METHODS Whole embryo culture model was used in this experiment.ICR mouse embryo(GD8.5)was contaminated with ethanol(4g/L)and intervened by nucleotide(0.16mg/L,4mg/L,100mg/L)for 48 h to evaluate the embryo development and morphological differentiation.RESULTS Supplementation with nucleotide at the dose of 0.16mg/L or 100mg/L significantly increased crown-rump length,visceral yolk sac,and scores of flexion and tail neural tube.4mg/L nucleotide supplementation significantly increased the scores of heart,hindbrain,midbrain,forebrain and otic,optic,olfactory system as well as forelimb and hindlimb in addition to these,compared with the negative group.CONCLUSION Nucleotide supplementation could resist the teratogenic action of ethanol in mice.
