摘　　要：目的探讨miRNA-29a在非小细胞肺癌(NSCLC)中的表达及临床意义。方法采用实时荧光定量PCR技术检测55例NSCLC组织及癌旁组织中miRNA-29a的表达水平,并分析其与临床病理特征及预后的关系。结果 NSCLC组织中miRNA-29a的表达下调43例,表达上调12例,其表达仅与性别有关(P<0.05),而与年龄、吸烟史、病理类型、肿瘤大小及TNM分期均无关。miRNA-29a表达诊断癌组织与癌旁组织的受试者工作特征(ROC)曲线的面积(Az)为0.669(95%CI:0.568～0.770),灵敏度和特异度分别为49.1%和85.5%。生存分析显示,不同miRNA-29a表达的NSCLC患者的生存率有明显差异(P=0.032);多因素回归分析显示,miRNA-29a表达是总生存期的独立预后因素。结论 miRNA-29a在NSCLC组织中表达下调,其可作为NSCLC的诊断、预后判断指标及治疗靶标。Objective To explore the expression of miRNA-29a in non-small cell lung cancer（NSCLC） and its relationship with clinical pathological features and prognosis. Methods The expressions of miRNA-29a in 55 NSCLC and adjacent cancerous tissues were tested by real-time PCR with U6 as internal control. Results NSCLC tissues were divided into 2 groups according to the expression levels of miRNA-29a, down-regulation group （n = 43 ） and up-regulation group（ n = 12）. miRNA-29a expression only correlated to gender,while didn＇t correlate with age, smoking history, histologic type, tumor size and TNM staging, miRNA-29a expression had value of diagnosis in NSCLC by ROC curve. Az of RDC curve was 0. 669 （95 % CI： 0, 568-0, 770）, and sensitivity and specificity were 49. 1% and 85.5%. There was a significant difference of overall survival down-regulation group and up-regulation group. Cox regression analysis showed expression of miRNA-29a was an independent prognosis factor of overall survival. Conclusion The expression of miRNA-29a is down-regulated in NSCLC. miRNA-29a may not only act as a signature of diagnosis and predictor of prognosis, but also as a therapeutic target in NSCLC,
