摘  要:目的:研究核糖核苷酸还原酶M2(RRM2)在结肠直肠癌组织的表达及对癌细胞增殖和迁徙的作用。方法:应用组织芯片免疫组化技术测组织中RRM2的表达。用siRNA转染RRM2高表达的细胞株HCT116以验证干扰效果,CCK-8法检测细胞增殖能力,Transwell实验评估转染后癌细胞迁移能力。结果:结肠直肠癌组织中RRM2的表达明显高于癌旁正常组织(P<0.05),且与浸润深度、分化程度、TNM分期均有相关性;在结直肠细胞株中,HCT116细胞RRM2在mRNA和蛋白水平表达量最高,用干扰效率最高的siRNA下调RRM2表达可以明显抑制HCT116细胞的增殖能力。在Transwell迁徙实验中,RRM2干扰组穿膜细胞数为(11±2)个,显著低于阴性对照组[(29±4)个]和空白对照组[(23±3)个](P<0.01)。结论:数据显示RRM2在结肠直肠癌组织中呈过表达,且在结肠直肠癌进展和预后发挥重要作用,其有望成为评估结肠直肠癌进展及预后的潜在指标。Objective To investigate the expression of ribonucleotide reductase M2 subunit (RRM2) in colorectal cancer and its influence on proliferation and migration of human colorectal cancer cells. Methods The expression of RRM2 was detected by tissue microarray using immunohistochemistry. SiRNA nligos targeting RRM2 were transfected into HCTll6 cell line which had high expression of RRM2. The proliferation was assayed by CCK-8 assay. Changes in migration of the selected cell line were evaluated by Transwell test. Results The expression of RRM2 in colorectal cancer tissue was significantly higher than adjacent normal tissue (P〈0.05). RRM2 expression level had correlation with invasion depth, differentiation and TNM stage. Colorectal cancer cell line HCT116 had the highest expression of RRM2 at both mRNA level and protein level. Transfection with the most efficient siRNA oligo targeting RRM2 could significantly suppressed the proliferation of HCT116. The Transwell test showed that the number of cells crossing through chambers in RRM2-siRNA group (11±2) was significantly lower than that in negative control group (29±4) and in blank control group (23±3), P〈0.01. Conclusions Our data suggests that ribonucleotide reductase M2 subunit is overexpression in tumor tissues of colorectal cancer. RRM2 may play an important role in the progression and prognosis of colorectal cancer, and can be a potential indicator to assess the prognosis of colorectal cancer.
