摘　　要：目的检测靶向端粒酶逆转录酶(hTERT)蛋白及mRNA在介入干预前后胃癌患者组织中的表达。方法选择胃癌患者50例,其中介入干预组30例,静脉化疗组20例,化疗药物选用5-氟脲嘧啶、亚叶酸钙及草酸铂,分别在干预前、干预后2周及4周采用免疫组化法及原位杂交法检测胃癌患者组织hTERT蛋白及mRNA表达水平,并选取10例正常胃组织作为对照。结果与干预前比较,静脉化疗组和介入干预组hTERT蛋白及mRNA水平均显著下降,且干预后4周与2周比较差异有统计学意义(P<0.05)。与静脉化疗组比较,介入干预组二者水平降低更为显著(F值分别为4.299和4.065,P<0.05)。结论 hTERT基因表达水平可作为评估胃癌干预疗效及预后的一个重要参考指标。Objective To investigate the expression of human telomerase reverse transcriptase（hTERT） protein and mRNA before and after interventional treatment through transcatheter arterial chemo-embolization in patients with gastric cancer.Methods Among 50 patients,30 patients（intervention treatment group） were treated with transcatheter arterial chemo-embolization,however,the other 20 patients（intravenous chemotherapy group） were treated with intravenous chemotherapeutic agents including 5-flurouracil,calcium folinate and oxaliplatin.The expression levels of hTERT protein and mRNA in 50 cases of gastric carcinoma tissues and 10 cases of normal gastric tissues（control group） before interventional treatment and at 2 week and 4 weeks after interventional treatment were detected by immunohistochemistry and in situ hybridization.Results As compared with those before interventional treatment,the expression levels of hTERT protein and mRNA in intravenous chemotherapy group and interventional treatment group were significantly decreased,furthermore,there were significant differences between 2 weeks and 4 weeks after interventional treatment（P0.05）.As compared with that in intravenous chemotherapy group,the decrease of expression levels of hTERT protein and mRNA in interventional treatment group was more obvious,and there was a significant difference between the two groups（P0.05）.Conclusion The expression levels of hTERT protein and mRNA are significantly decreased after intravenous chemotherapy and interventional treatment,moreover,the changes in interventional treatment group are more obvious,so the expression of hTERT protein and mRNA can be used as an important reference index for evaluating the efficiency of interventional treatment and the prognosis of patients with gastric carcinoma.
