摘  要:目的检测临床肺癌组织中解偶联蛋白2的表达与分布,并分析其与缺氧诱导因子1α在肺癌发生的关系。方法通过荧光定量PCR方法检测肺癌和癌旁正常组织中解偶联蛋白2与缺氧诱导因子1α的表达,同时应用免疫组织化学、Western blot方法检测解偶联蛋白2在肺癌和癌旁正常组织中的表达。结果荧光定量PCR方法检测,肺癌组织中解偶联蛋白2与缺氧诱导因子1αmRNA的表达约是癌旁正常组织的3倍,Western blot方法检测,解偶联蛋白2在肺癌组织中的表达量约是癌旁正常组织的2倍;免疫组织化学方法检测,解偶联蛋白2主要在细胞质中表达,在肺癌组织中的表达量明显高于癌旁正常组织,且癌细胞的恶化程度与解偶联蛋白2的含量呈正相关。结论解偶联蛋白2与缺氧诱导因子1α在人肺癌组织中均有高表达,但两者无明显相关性。Objective To explore the expression and distribution of UCP2 in clin ical lung cancer tissue,and to analyze the relationship between UCP2 and HIF1α. Methods The distribution of UCP2 in paired human lung cancer and peritumoral spe cimens was detected by immunohistochemistry,quantitative RT-PCR and Western blo t,respectively.The expression of HIF1αwas detected by quantitative RT-PCR.Res ults Quantitative RT-PCR analysis showed that the expression levels of UCP2 and HIF1αmRNA in cancer tissue was about 3 folds higher than those in tumor-adjac ent normal lung tissue.The positive rate of UCP2 expression was higher in lung c ancer patients with metastasis than in those without metastasis.Immunohistochemi stry showed positive staining for UCP2 in lung cancer and positive staining for UCP2 was rarely seen in peritumoral specimens.Conclusion The mRNA expression of UCP2 and HIF1αare both increased in human lung cancers,UCP2 mRNA levels showed no significant correlation with increased HIF1αmRNA and the level of expressio n appears to correlate with the degree of neoplastic changes.
