糖尿病模型建立的方法比较

摘　　要：目的探讨糖尿病动物模型的建立方法,并对其进行比较。方法用四氧嘧啶和链佐霉素分别单独和联合对小鼠腹腔注射,建立糖尿病动物模型,造模前小鼠高脂高糖喂养。小鼠分3组:一组单独用四氧嘧啶腹腔注射1次;二组用链佐霉素单独腹腔注射,小剂量多次;三组用链佐霉素单独小剂量多次腹腔注射后,再用四氧嘧啶半量腹腔注射1次;每组还设一个正常组作对照。结果一组、二组、三组造模前后血糖水平分别为:(6.17±0.49)mmol/L,(11.15±4.33)mmol/L;(6.08±0.70)mmol/L,(10.82±3.22)mmol/L;(5.49±2.85)mmol/L,(13.89±4.41)mmol/L,各组造模前后比较均有显著性差异(P均<0.01);总胆固醇(TC)除二组显著升高(P<0.05),其他两组无显著性差异(P均>0.05)。一至三组造模成功率分别为21.2%,25.0%,41.7%。结论链佐霉素联合使用四氧嘧啶可提高的造模成功率,单独使用四氧嘧啶造模成功率较低,单独使用链佐霉素成功率居中,而总胆固醇偏高。Objective It is to approach and compare the methods of establishing animal model of diabetes mellitus.Methods The animal model of diabetes mellitus were established with alloxan and streptozotocin separately and in combination on mouse peritoneal injection,before making model mice were confronted high-fat feeding.Mice were divided into three groups： Group A of separate with alloxan injected intraperitoneally;Group B with single intraperitoneal injection of streptozotocin,multiple low dose;Group C with multiple low dose streptozotocin alone after intraperitoneal injection of alloxan,and half a time of intraperitoneal injection;each group was also provided with a normal control groups.Results Group A,B and C before and after making model,sugar content as follows：（6.17±0.49）mmol/L and（11.15±4.33）mmol/L,（6.08±0.70）mmol/L and（10.82±3.22）mmol/L,（5.49±2.85）mmol/L and（13.89±4.41） mmol/L,there were significant differences in each group before and after（P0.01）.Total cholesterol（TC） was significant rose in Group B（P0.05） but was none in the others group.The success rate was 21.2%,25.0% and 41.7% in Group A,B and C respectively.Conclusion Streptozotocin combine alloxan may improve the modeling success rate,single use alloxan modeling success rate is low,use alone streptozotocin success rate is centered.
