摘 要:目的 研究红毛五加多糖对胃癌细胞的抑制作用及其作用机制。方法 检测细胞群体倍增时间以了解该多糖对胃癌细胞增殖的影响 ;使用流式细胞仪检测该多糖对抑癌基因 p5 3、Fas及Fas L蛋白表达的影响。 结果 红毛五加多糖 3个样品均使胃癌细胞群体倍增时间延长 ,且使抑癌基因 p5 3蛋白表达增高 ;且AGPⅡ还使抑癌基因Fas及Fas L蛋白表达增高。 结论 该多糖抑制胃癌细胞的增殖 ,其抑癌机制与抑癌基因 p5 3、Fas及Fas L蛋白表达有关。Objective In order to study the antitumor effects and their mechanisms of the Acanthopanax giraldii harms Var Hispidus Hoo polysaccharides(AGP) on the gastric cancer cell.Methods Cell double time was detected to reflect the proliferation of gastric cancer cell and flow cytometry was adopted to explore the protein expression of p53、Fas and Fas L.Results Cell double time was prolonged and protein expression of antioncogene p53 was high after the gastric cancer cell line SGC 7901 was treated with AGPⅠ、AGPⅡand AGPⅢ.Moreover,the protein expression of Fas and Fas L was high after the cells were cultured with AGPⅡ.Conclusion The AGP inhibited the proliferation of the gastric cancer cell and their mechanisms have some relationship with the high protein expression of p53、Fas and Fas L genes.
