摘 要:目的观察叶酸对人胃癌SGC-7901细胞增殖及ABCG2表达的影响。方法应用叶酸处理人胃腺癌SGC-7901细胞;MTT法检测细胞活力;免疫细胞化学SP法和RT-PCR分别检测ABCG2蛋白及mRNA的表达。结果(1)MTT法证实,与空白对照组相比,叶酸组SGC-7901细胞的增殖明显受抑制,差异有统计学意义(P<0.01),且随时间的延长和剂量的增加,胃癌SGC-7901抑制率呈上升趋势。(2)免疫细胞化学和RT-PCR结果分别显示,叶酸为低浓度时(10、50μg/ml),虽然随着浓度增加,ABCG2蛋白及mRNA表达有增强趋势,但与空白对照组相比,差异无统计学意义(P>0.05);当叶酸为高浓度时(250μg/ml),ABCG2蛋白及mRNA表达增加显著,差异有统计学意义(P<0.01)。结论叶酸对胃癌细胞存在抑制作用,但叶酸能促进ABCG2表达。表明叶酸对肿瘤具有一定的治疗作用,但高剂量叶酸可促使肿瘤发生耐药,导致肿瘤复发。Objective To observe the effects of folic acid on cell proliferation in human gastric cancer cell line SGC- 7901 and the expression of ABCG2. Methods SGC-7901 cells were treated in vitro with different concentrations of folic acid. There were the normal control group and the folic acid groups with different concentrations. The cells' proliferative vitality was de- tected by MTF. The expression of ABCG2 protein and ABCG2 mRNA was detected by immunoeytochemistry (ICC) and RT-PCR respectively. Results (1)The influence of folie acid on the growth of SGC-7901 in vitro:compared with the normal control group,folic acid at different concentrations groups could inhibit the growth of SGC-7901 significantly (P〈0.01). MTr conformed that the proliferation of SGC-7901 cancer cells was obviously inhibited by folic acid in a dose-and time-dependent manner, and the inhibition rate reached 50.72 % after treatment with 500 p,g/ml folic acid for 72 h. (2)ICC and RT-PCR respectively revealed that ABCG2 protein and mRNA increase in dose-dependent manner,but compared with untreated control cells,low folic acid (concentrations of 10,50 p^g/ml) could not make ABCG2 protein and ABCG2 mRNA increase significantly (P〉0.05). However in the high folic acid (250 p^g/ml) group ,ABCG2 protein and ABCG2 mRNA were increased significantly (P〈0.01). Conclusion Folie acid can effectively inhibit the proliferation of SGC-7901 human gastric cancer cell. But it can up-regulate significantly the expression of ABCG2 protein and ABCG2 mRNA. It makes clear that folic acid plays a part in tumor therapy. But high dose folic acid can result in drug-insistence and tumor recurrence.
