摘 要:目的观察新生大鼠脑组织缺血后血清基质金属蛋白酶-9(MMP9)、基质金属蛋白酶组织抑制物1(TIMP1)表达水平的变化。方法 75只7日龄Wistar大鼠分为正常对照组、缺血组、假手术组。采用左侧大脑中动脉结扎术建立脑缺血模型;对照组不做任何处理;假手术组只做一侧颈部正中切口,分离颈总动脉,不结扎。24小时后用酶联免疫吸附法(ELISA)检测3组大鼠血清MMP9、TIMP1表达水平。结果①脑缺血组血清MMP9水平(1.058±0.081)ng/ml,较对照组(0.993±0.090)ng/ml、假手术组(0.989±0.084)ng/ml明显升高,差异有显著的统计学意义(P<0.01);缺血组血清TIMP1水平(1.373±0.097)ng/ml,较对照组(1.294±0.074)ng/ml及假手术组(1.319±0.070)ng/ml亦明显升高,差异有显著的统计学意义(P<0.01)。②缺血组MMP9与TIMP1水平呈显著正相关(r=0.429,P<0.05);正常对照组和假手术组MMP9与TIMP之间无显著相关性(P>0.05)。结论 MMP9/TIMP1表达失调可能参与新生鼠缺血脑组织损伤。Objective To investigate the matrix metalloproteinase-9(MMP9)and tissue inhibitors of metalloproteinases 1(TIMP1)levels in newborn rat serum after cerebral ischemia. Methods Seventy-five 7-day-old rats were classified into 3 groups randomly:the control group,the ischemia group and the sham operation group.The rats in the ischemia group were subjected to right middle cerebral artery ligation.Rates in the sham operation group underwent incision on one side of the neck without ligation to separate the common carotid artery.The MMP9 and TIMP1 levels in serum were detected by enzyme linked immunosorbent assay 24 hours later. Results The serum MMP9 levels increased significantly in the ischemia group [(1.058 ± 0.081)ng/ml] compared with those in the control [(0.993 ± 0.090)ng/ml] and sham operation group [(1.319 ± 0.070)ng/ml](P 0.01).The serum TIMP1 levels increased significantly in the ischemia group [(1.373 ± 0.097)ng/ml] compared with those in the control [(1.294 ± 0.074)ng/ml] and sham operation group [(1.319 ± 0.070)ng/ml](P 0.01).The MMP9 and TIMP1 levels in serum showed positive correlation in ischemia group(r = 0.429,P 0.05).No correlation was founded in control and sham operation group(P 0.05). Conclusions The abnormal expression of MMP9 or TIMP1 may contribute to ischemic brain injury newborn rats.
