摘  要:目的:观察热敏灸对抑郁症模型大鼠学习记忆力及海马脑源性神经营养因子(BDNF)mRNA表达的影响,研究热敏灸治疗抑郁症的效果及作用机制。方法:雌性SD大鼠,采用孤养和慢性不可预见性温和应激(CUMS)制备抑郁症大鼠模型。以Morris水迷宫测试大鼠的学习记忆力;以RT-PCR技术检测大鼠海马BDNF mRNA的表达量。结果:模型组大鼠学习记忆力下降,海马BDNF mRNA的相对表达量减少;与模型组相比,热敏灸组大鼠学习记忆力改善,海马BDNF mRNA的相对表达量增加。结论:热敏灸具有改善抑郁症大鼠模型的学习记忆力,可能与其提高海马BDNF mR-NA的表达有关。Objective : The effect of heat-sensitive moxibustion on spatial learning-memory abilities and the relative expression of brain-derived neurotrophic factor(BDNF) mRNA in hippocampus were investigated in order to explore the mechanism of its an- tidepressant effect in depression model rats. Methods :Twenty-seven female SD rats were randomly divided into four groups:normal group, model group,moxibustion group and heat-sensitive moxibustion group. Depression model was induced by giving isolationhousing in combination with chronic unexpected mild stress(CUMS) for 21 days. Morris water maze was used to test spatial learn- ing-memory abilities. Reverse transcription-polymerase chain reaction(RT-PCR) was employed to detect the relative expression of BDNF mRNA. Results : Compared with rats in normal group, rats in model group showed that spatial learning-memory abilities and the relative expression of BDNF mRNA in hippocampus declined. And compared with model group rats, heat-sensitive moxibustion group rats showed improving spatial learning-memory abilities and increasing the relative expression of BDNF mRNA in hippocam- pus. Conclusion:The heat-sensitive moxibustion can improve spatial learning-memory abilities, which might be mediated through increasing the relative expression of BDNF mRNA in hippocampus of depression model rats.
