摘  要:目的探讨健康人群NK细胞在体外培养的最佳时间。方法抽取健康人群外周血15ml,用常规单个核细胞提取方法提取单个核细胞,放于含有SCGM培养基的培养孔中培养,前5d加入0.5μlCD3McAb,在培养的第5d,倒掉含有CD3McAb的培养液,重新加入含有rhIL-2的SCGM培养基。隔日补充1次培养液,直至第25d。同时分别检测培养第0d、5d、10d、15d、20d、25d培养细胞的细胞数量、细胞存活率、NK细胞的百分率、T细胞的百分率及NK细胞对K562细胞和RPIM8226细胞的杀伤活性。结果培养前20d,总细胞和NK细胞的数量、NK细胞的百分率、NK细胞对K562细胞和RPMI8226细胞的杀伤活性与培养时间呈正相关;T细胞的百分率与培养时间呈负相关。结论健康人群NK细胞的体外最佳培养时间是20d。Objective To investigate the optimal culture time of the healthy population's NK ceils. Methods Extracting the mononuclear cells in peripheral blood 15ml of 10 healthy individual with conventional mononuclear cell extraction method, and the mononuclear ceils were put in the cultivation hole containing SCGM medium, for the first 5 days, the medium was supplemented with 0.5μl CD3McAb, on day 5 of culture, the CD3McAb-containing medium was washed out, and fresh medium with rhIL-2 was added. The cultures were then replenished with fresh medium every other day on to the day 25. Determing the cell number, cell viability, percentage of NK cells and T cells, the killing activity of elected NK cells against K562 and RPMI8226 ceils on days 0, 5, 10, 15, 20, 25 of culture. Results On the first 20 days, The number of total cells and NK cells, the percentage of NK cells, the killing activity of NK cells against K562 ceils and RPMI8226 ceils all are positivly correlated with culture time, the percentage of T cells is negatively correlated with culture time. Conclusion The optimal culture' time interval of the healthy population's NK cells is 20 days.
