摘 要:目的探讨以胶原蛋白海绵为载体,复合重组人骨形态发生蛋白-2(rhBMP-2)植入兔耳软骨损体内诱导软骨膜再生软骨的效果。方法选用日本大耳白兔9只,随机将兔分为3组,分别完整去除1 cm×1cm软骨,完整保留软骨膜。A组植入胶原蛋白海绵组、B组植入含0.5mg/ml的rhBMP-2/胶原蛋白海绵复合物、C组不植入任何材料。术后4、8、12周分批处死动物,取材,去除软骨膜观察新生软骨情况、面积;通过HE染色、Masson染色和醛品红染色,观察标本中细胞浸润和分化生长情况、新生软骨情况和弹力纤维变化和生成情况。采用HPIAS-1000高清晰度彩色病理图像分析仪测定新生软骨的面积。结果组织学评分A组、B组和C组在各个时期均存在显著性差异,有统计学意义(P<0.01)。两个实验A、B组4、8、12周比较差异有统计学意义(P<0.01),同一时间点两实验组间比较差异均无统计学意义(P>0.05)。新生软骨面积,胶原蛋白海绵组、rhBMP-2/胶原蛋白海绵复合物组、空白对照组像素分别为1452±549.8、1962±524.4、1197±496.2。方差分析,三组结果差异有统计学意义(P<0.05)。结论 rhBMP-2能促进促进软骨膜处的软骨再生。胶原蛋白海绵可作为rhBMP-2安全有效的载体,同时为软骨膜再生软骨提供条件,二者复合体的应用为软骨切取术后的修复软骨缺损提供了一种新方法。提示胶原蛋白海绵有利于软骨膜再生的作用。Objective The discussion to the collagen sponge as a carrier, combined with recombinant human bone morphogenetic protein 2(rhBMP-2) implantation in rabbit ear cartilage defects in vivo induced cartilage membrane cartilage regeneration effect. Methods Selection of 9 lapanese rabbits, rabbits were divided into 3 groups randomly, respectively, complete removal 1cm×1cm cartilage, intact cartilage membrane. Group A implantation of collagen sponge group, B group of implanted with 0.5mg/ml rhBMP-2/collagen sponge composite, group C implanted in any material. Besides, area, by HE staining, Masson staining and aldehyde fuchsin staining, specimens were observed in ceil invasion and differentiation of cartilage growth, and elastic fiber changes and formation. Using HPIAS-1000high definition color pathological image analyzer for the determination of cartilage surface Product. Results Histological grading, in A group, B group and C group in each period, there were significant differences were statistically significant(P〈0.01). In two experiments A, B group 4,8,12 weeks had a significant difference(P〈0.01), at the same time in two experimental groups showed no significant differences(P〉0.05). The area of new cartilage Collagen sponge group, rhBMP-2/ collagen sponge complex group of pixels of blank control group were 1452±549.8, 1962±524.4, 1197±496.2. The analysis of variance, the three groups resulting in significant difference was statistically significant(P〈0.05). Conclusion rhBMP-2 can promote to promote cartilage at the cartilage regeneration. Collagen sponges as rhBMP-2 is safe and effective carrier, at the same time provide the conditions for cartilage regeneration of cartilage. The application of the two complexes for cartilage cut after surgery to repair the cartilage defect to provide a new approach, suggesting that the collagen sponge to the role of cartilage regeneration.
