摘 要:目的观察苦参碱以及苦参碱联合顺铂对小鼠宫颈癌细胞株u14移植瘤中肺癌抑制因子1(TSLC1)蛋白表达的影响。方法通过接种小鼠宫颈癌细胞株u14建立荷瘤小鼠模型,随机分为组:阴性对照组(给予生理盐水),阳性治疗组(给予顺铂2mg.kg-1),苦参碱高、中、低剂量组(分别给予75mg.kg-1、50mg.kg-1、35mg.kg-1的苦参碱),苦参碱高、中、低剂量组分别联合2mg.kg-1顺铂治疗组,均采用腹腔注射,用药10天后,采用脱臼处死小鼠并剥取肿瘤组织;通过HE染色观察移植瘤组织细胞形态变化,免疫组化方法观察移植瘤组织中TSLC1蛋白的表达情况。结果 HE染色结果显示移植瘤组织呈现典型的鳞状细胞癌的表现;免疫组化结果显示,随着苦参碱剂量的增加,移植瘤组织中的TSLC1蛋白的阳性表达率逐渐上升,苦参碱高、中剂量组显著高于阴性对照组(P<0.01);高、中、低剂量的苦参碱联合2mg.kg-1顺铂治疗后TSLC1蛋白阳性表达率分别为90%、80%、70%,与阴性对照组比较均具有显著统计学意义(P<0.01)。结论苦参碱具有上调u14移植瘤组织中TSLC1蛋白表达的作用,可能是苦参碱抗宫颈癌的重要作用机制之一。Objective To observe the impact of Matrine and DDP on the expression of TSLC1 in a mouse tumor model of u14 cervical cancer.Methods Mice tumor models of u14 cervical cancer were established by inoculating mouse carcinoma cell of cervix uteri line U14 and divided randomly into following groups:negative group(treated by saline),positive group(treated by DDP),matrine groups(35 mg·kg^-1,50 mg·kg^-1 and 75 mg·kg^-1);matrine plus DDP groups(treated respectively by different doses of matrine as well as 2 mg·kg^-1 DDP).All mice were sacrificed after treatment for 10 days,the morphological change of transplantable tumors were detected under microscope by HE staining,the expression of TSLC1 protein in u14 transplantable tumors were detected through immunohistochenmistry Results The HE staining under the microscope showed that the tissue of u14 transplantable tumors were typical squamous cell carcinoma;with the increase of dose of matrine,the positive rates of TSLC1 in u14 transplantable tumors gradually rised;positive rates in the groups that were treated by matrine at doses of 50 mg·kg^-1 and 75 mg·kg^-1 were significantly higher than the negative group(P〈0.01);the positive rates of TSLC1 in the groups which were treated by matrine as well as 2 mg·kg^-1 DDP were respectively 90%,80% and 70%,all were significantly higher than the negative group(P〈0.01).Conclusion Matrine possessed the ability to upregulate the expression of TSLC1 protein in u14 transplantable tumors,which may be one of the important mechanisms of the effects of matrine against cervical cancer.
