摘  要:目的观察骨髓间充质干细胞(BMSCs)移植对电离辐射诱发的小鼠胸腺瘤血管内皮生长因子(VEGF)表达的影响,为探讨BMSCs在辐射致癌中的作用奠定基础。方法采用X射线照射C57BL/6小鼠建立胸腺淋巴瘤模型,HE染色观察胸腺组织的病理变化,并判断成瘤情况。将C57BL/6小鼠随机分成正常对照组、照射组及照射+MSCs组。6个月后取胸腺组织,提取总RNA、合成cDNA,采用RT-PCR对VEGF基因mRNA进行检测。结果 C57BL/6小鼠经照射后,胸腺组织皮质和髓质分界不清,淋巴样肿瘤细胞弥漫分布,侵犯周围的脂肪组织,形成多泡型。胸腺瘤VEGF基因mRNA表达水平(1.3280±0.2385)高于正常胸腺组织(0.8263±0.3844)和BMSCs输注组(0.8931±0.2268),差异有统计学意义(P﹤0.05)。结论成功建立辐射诱发的小鼠胸腺瘤模型;VEGF基因与辐射诱发小鼠胸腺瘤密切相关,输注的MSCs可下调VEGF基因的表达,降低胸腺瘤的成瘤率。OBJECTIVE To study the influence of bone marrow mesenchymal stem cells on the expression of VEGF mRNA on thymoma induced ionizing radiation in mice, and to to provide a theoretical basis for the role of BMSCs in the process of tumor development. METHODS The model of Thymoma induced by whole-body X-irradiation was established. Pathological change was observed by HE staining, and the thymoma formation was judged. The mice were divided into three groups: the normal, irradiation and treatment groups with MSCs after irradiation. VEGF gene mRNA expression was detected by RT-PCR 6 month later. RESULTS The structures of cortices and medulla of thymoma induced ionizing radiation was unclear, lymphoid tumor cells diffused, and violated the surrounding fat tissue. The VEGF mRNA expression of thymoma (1.328 0 ± 0.238 5) was sig- nificantly higher than that of normal thymus tissue (0.826 3 ± 0.384 4) and MSCs group (0.893 1 ± 0.226 8) (P﹤0.05). CONCLUSION The model of thymoma induced by ionizing radiation was successfully established. MSCs can inhibit tumor proliferation through regulating the expression of VEGF.
