摘  要:目的评价改进后的线栓法大鼠永久性大脑中动脉阻塞局灶脑缺血模型(PMCAO)的使用价值。方法 SD大鼠随机分为3组,即正常对照组(n=8)、假手术组(n=8)、脑缺血组(n=16);脑缺血组分别取术后6 h、24 h为观察点,每个时间点各8只大鼠。通过改进颈部血管暴露方法、线栓进入技巧等实验技术,线栓法建立PMCAO后,2%红四氮唑溶液(TTC)染色观察各组缺血侧脑梗死变化和苏木素—伊红(H-E)染色观察各组缺血侧病理改变。结果脑缺血组术后6 h、24 h脑缺血侧脑梗死和脑组织病理改变随时间加剧,而正常对照组和假手术组未见脑梗死和相应的病理改变。造模成功率达90%。结论改进后的线栓法PMCAO方法操作简单,结果稳定可靠,重复性好,脑梗死与临床病理过程一致,是研究局灶脑缺血损伤比较理想、简易的实验动物模型。Objective To study the worth of focal cerebral ischemia model of permanent middle cerebral artery oc- clusion (PMCAO) induced by suture method improved from the classic method. Methods SD rats were randomly divid- ed into three groups : normal control group ( n = 8 ) ; sham-operated group ( n = 8 ) ; cerebral ischemia group, in which the rats were divided into 6 h or 24 h cerebral ischemia group (n = 8 for each). Cerebral infarction area was assessed by staining with 2% 2,3,5-triphenyltetrazolium chloride (TYC) solution and pathological changes of cerebral infarction was assessed by staining with hematoxylin-eosin (H-E). Results Cerebral infarction area continuously increased and pathological changes of cerebral infarction continuously exacerbated at ischemic hemisphere from 6 h to 24 h after middle cerebral artery occlusion, while similar changes did not found in normal control and sham-operated rats. In additional, to use this method, 90% of them is successful. Conclusion The modified PMCAO model can be operated simply and is reliable, stable and repeatable, and cerebral infarction is consistent with the clinical pathological process. It is perfect PMCAO model to study fo- cal cerebral ischemic injury.
