SOCS3在皮肤黑素瘤中的表达及意义

摘　　要：目的:探讨SOCS3蛋白在皮肤黑素瘤和色素痣组织中的表达及意义。方法:应用免疫组织化学S-P法检测90例皮肤黑素瘤和43例色素痣组织中SOCS3、基质金属蛋白酶-2(matrix metalloproteinases,MMP-2)、MMP-9的表达情况,分析它们与患者临床病理特征的关系。结果:SOCS3蛋白在皮肤黑素瘤组织中的表达明显低于色素痣(分别为40.0%和60.5%,P<0.05)。SOCS3低表达与肿瘤的厚度、侵袭深度、淋巴结转移均有一定关系(P<0.05～P<0.01);黑素瘤患者SOCS3表达阳性者的无复发生存和总生存率均较阴性者有显著性提高(P<0.01)。此外,SOCS3表达与MMP-2和MMP-9(P<0.01)蛋白表达均呈负相关关系(P<0.05和P<0.01)。结论:皮肤黑素瘤SOCS3表达与肿瘤生长、侵袭转移和预后密切相关,提示SOCS3蛋白可能在皮肤黑素瘤的侵袭生长过程中发挥了重要作用。Objective ：To study the expression and significance of suppressor of cytokine signaling 3 （SOCS3） in cutaneous melanoma and nevus tissues. Methods： Immunohistochemical S-P method was used to detect the protein expression of SOCS3, mat/ix metalloproteinases-2 （MMP-2） and MMP-9 of 90 cases with melanoma and 43 cases with nevus, and analysed the relation with the clinical and pathologic features of all patients. Results ：The positive expression of SOCS3 in melanoma was significantly lower than that of nevus tissues （40% and 60.5% respectively, P 〈 0.05 ） , and. The low expression of SOCS3 was correlated to the diameters and invasive depth of tumors and metastasis of lymph node（ P 〈 0.05 to P 〈 0.01 ）. Compared with the cases with negative expression of SOCS3 ,the cases with melanoma with positive SOCS3 expression had significantly longer relapse-free survival and overall survival（P 〈 0.01 ）. SOCS3 expression was inversely correlated with the expression of MMP-2 and MMP-9 （P 〈 0.05 and P 〈 0.01 ）. Conclusions： The expression of SOCS3 was closely related to the tumor growth, invasion and metastasis, SOCS3 may play an important role on the invasive growth of human cutaneous melanoma.
