摘  要:采用(NH4)2SO4为无机氮源,利用实验室筛选的高产魔芋葡甘聚糖酶的B23菌种,在50 L发酵罐中进行中试放大,研究了工业发酵生产中的生产条件以及pH的调控,讨论了工业生产魔芋葡甘聚糖酶的可行性。结果表明:pH值7.0、接种量6%、通气量0.8 m3/h、搅拌速度100 r/min为最佳条件,总酶活力达到2 574 U/mL。优化了从实验室摇瓶生产至发酵罐生产中必须测定的通气量、搅拌速度、接种量等工业生产条件,提高了该菌在实验室摇瓶发酵的酶活力,可进入工业生产研究。The(NH4)2SO4 was taken as the inorganic nitrogen source,the B23 bacterium,a high konjac glucomannanase producing strain,was taken to conduct pilot production in 50 L of fermentor in order to study the producing conditions and pH regulation and control during commercial fermenting production and to discuss the feasibility of commercial producing konjac glucomannanase of Amorphophallus rivieri Durieu.The results showed that the best fermentation conditions were inoculum size of 6%,aeration rate of 0.8 m3/h and stirring speed of 100 r/m.Under the best conditions,the total enzyme activity reached 2 574 U/ml.The aeration rate,stirring speed and inoculum size,which must be determined during the changing process from laboratory flask-shaking production to fermentor production,were optimized,and the activity of B23 in laboratory flask-shaking production were enhanced.Therefore,the trial-production can be performed.
