人主动脉内皮细胞的培养及免疫细胞化学研究

摘　　要：人主动脉内皮细胞体外长期培养在国内首次成功。内皮细胞培养10～15代,细胞倍增时间20h～25h,累积倍增数25～30。应用相差显微镜,扫描、透射电镜,Ⅷ因子相关表面抗原的免疫荧光抗体及血管紧张素转换酶的单克隆抗体,对培养的人主动脉内皮细胞进行形态学及免疫细胞化学的观察与研究。证明,长期培养后的多核内皮细胞是具有内皮细胞特点的变形内皮细胞。We successfully cultured endothelial cells from human aorta first at home.Cells survied 10-15 passages (doub1e time of cells 20-25 hours,25-30 cumulativepopulation doublings). These long-term cultured endothelial cells were observedmorphologically under phase-contrast microscope, scanning, transmission electronmicroscope and investigated immunocytochemically by immunofluorescence ofspecific antibody against Factor-Ⅷ related surface antigen and ABC methedusing monoclonal antibody 9B9 against human angiotensin-converting enzyme.The large multinuclear endothelial cells presented in long-term culture are vari-ant endothelial cells.
