摘 要:为探究雌激素与抗雌激素的复合效应,将斑马鱼(Danio rerio)的胚胎暴露于o,p'-DDT和氟维司群2种典型的环境内分泌干扰物中,考察胚胎孵化率、孵化时间和总畸形率,并用实时荧光定量PCR技术检测Vtg1、Vtg2和ERα等雌激素效应相关基因的mRNA转录水平。结果显示,与单一化合物的暴露相比,o,p'-DDT和氟维司群的二元混合物暴露对斑马鱼的发育毒性增强。在诱导Vtg1和ERα基因的mRNA表达水平上调方面,氟维司群对o,p'-DDT的诱导效应表现出拮抗作用,而当50或250μg·L-1氟维司群与100μg·L-1o,p'-DDT共存时,对Vtg2有明显的诱导效应。研究表明,抗雌激素并非简单地抑制环境雌激素的生态毒性效应,在进行生态风险评价时需全面评估雌激素与抗雌激素的复合效应。Abstract: To study the combination effects of estrogen and antiestrogen, zebrafish (Danio rerio) embryos were exposed to the typical estrogen o, p'-DDT and antiestrogen fulvestrant, and the hatching rate, hatching time and gross abnormality of embryos were investigated. Besides, real-time quantitative PCR was used to probe the rnRNA expression levels of estrogen-responsive genes such as Vtgl, Vtg2 and ERa. Results showed that co-treatment with o, p'-DDT and fulvestrant led to an increase in developmental toxicity to ze- brafish embryos, compared with single compound exposure. The expressions of Vtgl and ERa induced by o,p'-DDT were inhibited by fulvestrant. However, the expression of Vtg2 was found to be significantly in- duced, when embryos were co-treated with fulvestrant (50 or 250 Ixg'L 1) and o,p'-DDT (100 Ixg'L-1). It is demonstrated that the effects of antiestrogen could not be simply summarized as neutralization in the combi- nation exposure of estrogen and antiestrogen. It is critical effects when conducting ecological risk assessment. to get a comprehensive view of the combination
