摘 要:HL-60细胞在DMSO诱导下沿粒细胞方向分化,120h NBT^+细胞达84%。细胞经诱导24h蛋白激酶C活性增加,72h为未诱导细胞的2.7倍。酶活性的增加,主要是由于细胞浆部分而不是细胞膜部分可溶性酶活性改变的结果。分化诱导剂DMSO本身不能直接激活蛋白激酶C,提示DMSO对酶活性的影响是间接的。Treatment of HL-60 cells with DMSO caused cells to differentiate along the myeloid way and 84% of the cells showed a reduction in NET after 120h. Protein kinase C activity increased of during differentiation. It would reach 2.7 times that of the control after 72h in the presence of DMSO. The increase of protein kinase C activity was traced to the cytosolic rather than the membrane fraction. Chelator EGTA was necessary to extract protein kinase C. DMSO itself could not enhance protein kinase C activity, indicating that the effect of DMSO was indirect. All of these results showed that the activation of protein kinase C is important in the differentiation of HL-60 cells.
