葫芦素B和阿霉素联合应用在体内外对肝癌细胞H22的生长抑制作用

摘　　要：目的研究葫芦素B和阿霉素联合应用在体内外对肝癌细胞H22的生长抑制作用。方法将40只雌性昆明小鼠随机分为4组,每组10只,包括阴性对照组(生理盐水组)、阿霉素组、葫芦素B组、联合组。分别以阿霉素、葫芦素B、葫芦素B联合阿霉素处理H22细胞,培养48 h后MTT法检测细胞增殖情况。计算抑瘤率、白细胞数、胸腺指数、脾脏指数和血药浓度。结果 MTT结果显示,与阿霉素组相比,联合组对H22细胞的增殖抑制作用明显增强,联合组与阿霉素组之间、高剂量组与低剂量组之间的差异均有统计学意义(P<0.05或P<0.01)。联合组与阿霉素组相比,抑瘤率明显增强,并且联合组阿霉素在心脏中的浓度明显降低(P<0.05)。结论葫芦素B和阿霉素联合应用具有增强抗H22肝癌作用。Objective To investigate the effect of cucurbitacin（CuB）combined with doxorubicin（Dox）on hepatocellular carcinoma cells.Methods 40 female Kunming mice were randomly divided into negative control group（saline group）,Dox group,CuB group and combination group,10 mice in each group.H22 cells were treated with Dox,CuB,and both CuB and Dox.The proliferation of cells was tested by MTT assay after 48 h,and the rates of restraining tumor,leukocyte count,thymus index,spleen index and plasma concentration were calculated.Results Compared with the Dox group,inhibition on H22 cell proliferation in combination group was significantly enhanced,and there were significant differences between combination group and control group,high dose group and low dose group（P〈0.05 or P〈0.01）.Compared with the control group in combination group,the tumor weight decreased,and the Dox concentration in heart was lowered significantly（P〈0.05）.Conclusion The combination of CuB with Dox shows a synergistic effect on H22 murine hepatocellular carcinoma.
