摘　　要：目的:研究膜型基质金属蛋白酶1(membranetype1matrixmetalloproteinase,MT1MMP)基因mRNA在人胃癌组织中的表达及其与胃癌浸润转移和临床分期的关系。方法:应用RTPCR半定量的方法检测MT1MMP基因mRNA在30例胃癌组织、正常粘膜和转移淋巴结中的表达。使用SPSS/PC+统计软件进行t检验和多因素分析。结果:MT1MMP基因的mRNA在肿瘤组织和转移淋巴结中分别有76.7%(23/30)和76.9%(10/13),高于正常组织。按TNM分期,MT1MMP在Ⅰ、Ⅱ期与Ⅲ、Ⅳ期的表达差异有显著性。经多元回归分析,MT1MMP基因的表达与肿瘤的浸润深度、有无淋巴结转移及有无癌栓正相关。结论:MT1MMP基因参与胃癌的直接浸润和转移,可能成为一种潜在的肿瘤生物学标记物。Objective: To study the expression of membrane type-1 matrix metalloproteinase(MT1 MMP) in human gastric carcinomas and the relationship between the expression and tumor invasion, metastasis, and TNM classification. Methods: By using RT PCR to determine the expression of MT1 MMP mRNA in 30 pair cases of gastric carcinoma, corresponding normal mucosa and partial metastasis lymphoden. The result was assessed with t test and multiple regression using SPSS/PC+ statistical software. Results: The expression level of MT1 MMP mRNA of the tumor was greater than that of the normal mucosa in 23 of the 30 cases(76.7%). The expression in lymphodus was greater than that in the corresponding normal mucosa in 10 of the 13 cases(76.9%). The expression of MT1 MMP in stages Ⅰ,Ⅱ and that in stages Ⅲ,Ⅳ (TNM classification)had significant difference By multiple regression, the expression of MT1 MMP was positively correlated with lymphodus metastasis, invasive depth and tumor embolism. Conclusion: These findings suggest that the overexpression of MT1 MMP may play an important part in tumor direct invasion and metastasis, MT1 MMP may also prove to be a useful marker for determing the biological aggressiveness of gastric carcinoma.
