脑膜瘤相关蛋白、环氧化酶2蛋白在食管鳞癌中的表达意义及相关性研究

摘　　要：目的探讨食管鳞癌及正常食管黏膜中脑膜瘤相关蛋白(meningioma associated protein,MAC30)、环氧化酶2(cyclooxygenase-2,COX-2)蛋白的表达及相关性,并分析其与食管鳞癌临床病理特征的关系。方法采用免疫组织化学链霉菌抗生物素蛋白-过氧化物酶连结(SP)法检测68例食管鳞癌及22例正常食管黏膜中MAC30、COX-2表达情况。结果 MAC30、COX-2在食管鳞癌中的表达分别明显高于正常食管黏膜,50.0%vs 4.5%,48.5%vs9.1%(P<0.01),同时在临床病理分期、淋巴结转移各组间差异有统计学意义(P<0.05),二者在食管鳞癌中的表达呈中等正相关(rs=0.324,P<0.05)。这两种蛋白表达与患者性别、年龄及肿瘤大体形态、分化程度均无相关性(P>0.05)。结论 MAC30、COX-2蛋白在食管鳞癌发生、发展中起关键作用,与其侵袭转移密切相关。食管鳞癌中MAC30蛋白与COX-2蛋白的表达呈正相关。Objective To study the expression and relationship of meningioma associated protein 30（MAC30）, cyclooxygenase 2 （COX-2） proteins in esophageal squamous cell carcinoma （ESCC） and normal esophageal mucous membrane tissue,and to analyze the correlation of MAC30 and COX-2 expressions with the clinicopathological features in ESCC. Methods MAC30, COX-2 expressions in 68 cases of ESCC and 22 cases of normal esophageal mucous membrane tissue were detected with immunohistochemical SP method. Results The expressions of MAC30, COX-2 proteins in ESCC were significantly higher than those of normal esophageal mucous membrane tissue,50.0% vs 4.5%, 48.5% vs 9.1%（ P 〈0.01） ,and were related to the clinical stage and lymph node metastasis （ P 〈0.05）. There was moderately positive correlation between MAC30 and COX-2（ r, =0. 324, P 〈0.05）. The expressions of the two proteins were not correlated with patients＇ gender, age, and the macroscopical shape and the degree of ESCC differentiation （ P 〉0.05）. Conclusion MAC30 and COX-2 may play key roles in carcinogenesis and development of ESCC,and may be closely related to invasion and metastasis of ESCC. There was positive correlation between the expressions of MAC30 and COX-2 proteins in ESCC.
