摘  要:目的:研究三七素对谷氨酸损伤的PC12细胞的影响。方法:用谷氨酸复制体外培养的PC12细胞损伤模型,采用MTT法、Hoechst33342/PI双重染色法分别研究高、低剂量三七素对谷氨酸所致的PC12细胞损伤的影响。结果:谷氨酸刺激后,PC12细胞存活率较对照组显著降低(P<0.05)、凋亡显著增强(P<0.05)。给予高剂量三七素可加重谷氨酸引起的PC12增殖力降低、凋亡增强。但低剂量三七素干预后,细胞存活率较模型组显著升高(P<0.05),凋亡较模型组明显降低(P<0.05)。结论:高剂量三七素可加重谷氨酸对PC12细胞的损伤,但低剂量三七素可显著减轻谷氨酸对PC12细胞的损伤,但具体机制尚有待于进一步研究。Objective: To investigate the effect of dencichine on PC12 cells injuried by glutamate. Methods: PC12 cells were injuried by glutamate, and then the cells were theraped respectively by dencichine at the doses of 0.5 mg/kg and 0.5 mg/kg. The proliferative ability and apoptosis of PC12 cells were detected by MTT and Hoechst33342/PI. Results: Dencichine at the dose of 2.5 mg/kg aggravated the inhibition of proliferative ability and apoptosis of PC 12 cells induced by glutamate, but dencichine at the dose of 0.5 mg/kg ameliorated the inhibition of proliferative ability and apoptosis of PC12 cells induced by glutamate. Conclusions: Dencichine is neurotoxic, dencichine and low dose of which could ameliorated the impairment caused by glutamate.
