摘  要:目的探讨X染色体耦联锌指蛋白(Zfx)在人脑胶质瘤干祖细胞中的表达及意义。方法逆转录PCR方法检测人脑胶质瘤干祖细胞SU2及其分化后贴壁细胞中mRNA的表达;细胞免疫组化检测人脑胶质瘤干祖细胞SU2中Zfx、CD133、nestin的表达;然后在含有10%胎牛血清培养液中诱导SU2分化,并检测分化后的细胞中Zfx、S100、GFAP的表达。结果 Zfx在胶质瘤干祖细胞SU2及其分化后贴壁细胞中的mRNA均有表达。Zfx在人脑胶质瘤干祖细胞SU2中的蛋白水平表达高,但SU2经诱导分化后细胞中Zfx蛋白水平表达减弱。结论 Zfx在人脑胶质瘤干祖细胞中高表达,可能在人脑胶质瘤干祖细胞自我更新和分化抑制过程中起重要作用。Objective To investigate the expression and significance of X-linked zinc-finger protein(Zfx) in human glioma stem cells. Methods The expression of Zfx mRNA in glioma stem/ progenitor cells SU2 and serum-induced cells was detected by RT-PCR The expressions of Zfx, CD133, nestin were examined by immunofluorescence staining. SU2 was cultured in medium supplemented with 10% FBS and induced to differentiate, in which The expressions of Zfx, S100, GFAP were examined by immunofluorescence stainings. Results Zfx mRNA was expressed in human glioma stem/progenitor cells and its differentiated cells. The expression of Zfx protein level was higher in brain glioma stem/progenitor cells than that in the differentiated cells. Conclusion Zfx is overexpressed in glioma stem/progenitor cells, which may play an important role in the self-renewal of glioma stem/progenitor cells.
