摘  要:目的研究弹性蛋白酶诱导兔颈内动脉瘤模型的建立及其瘤壁平滑肌细胞表型变化。方法应用弹性蛋白酶作用于兔颈内动脉,建立新西兰兔颈内动脉瘤模型;分别于第1、2、3、4周取下实验动脉瘤标本行HE染色,逆转录PCR检测瘤壁平滑肌细胞表型基因(SM22α-mRNA)表达。结果实验动脉瘤随着不断生长瘤壁逐渐变薄,弹性纤维断裂,平滑肌细胞减少,SM22α-mRNA表达减少。结论弹性蛋白酶能稳定诱导动脉瘤的产生。动脉瘤动态生长过程中血管平滑肌细胞表型由收缩型向合成型转化。Objective To establish the rabbit aneurysm model and observe the changes of vascular smooth muscle cells (VSMC) in aneurysm wall. Methods The carotid arterial aneurysm models of New Zeland rabbits were induced by exposure to elastase. The expression of SM22α-mRNA in VSMC of aneurysm wall was detected with histological HE staining and RT-PCR analysis. Results During growth of the aneurysm from 1 to 4 weeks, the vascular media became thinner and elastic fibres became fragmented, and the expression of SM22α-mRNA was decreased. Conclusion Elastase can stablly induce an aneurysm model. During the progress of aneurysm developing, VSMC is remodeled from the contractile phenotype to a synthetic phenotype.
