摘 要:目的:通过观察莪术油对大鼠乳腺癌癌前病变组织中血管内皮生长因子(VEGF)mRNA表达的影响,探讨莪术油治疗乳腺癌癌前病变的机制。方法:275只SD大鼠随机分为空白对照组、造模对照组、他莫昔芬组、康莱特组及莪术油小、中、大剂量组。采用二甲基苯葸(DMBA)诱导乳腺癌癌前病变造模,干预治疗4周,第8~14周分4批处死动物,进行病理学检测,原位杂交法测定标本中VEGF mRNA表达。结果:各试验组不同类型乳腺组织中VEGF mRNA表达阳性率及表达强度呈递增趋势。大鼠非典型增生乳腺组织中,莪术油小剂量组VEGF mRNA表达强度为(36.27±5.36)%,中剂量组为(30.83±4.12)%,大剂量组为(26.14±3.17)%,康莱特对照组为(45.49±2.86)%,他莫昔芬对照组为(47.43±6.32)%,造模对照组为(78.65±6.28)%。各干预组的VEGF mRNA表达强度均明显低于造模对照组(P<0.05),莪术油组低于康莱特和他莫昔芬组(P<0.05)。对VEGF mRNA表达阳性的非典型增生标本,莪术油大剂量组VEGF mRNA表达强度明显低于小剂量组,P<0.05。结论:莪术油能够有效的降低DMBA诱导大鼠乳腺癌癌前病变组织中VEGF mRNA表达强度,可能是其抑制血管生成、阻断乳腺癌发生的有效机制。OBJECTIVE:To research the treatment mechanism of zedoray turmeric oil (ZTO) in precancerous lesion of rats mammary by measuring the expression of VEGF mRNA. METHODS: Totally 275 SD rats were randomly divided into 7 groups:the blankcontrol group (BC), the precancerous lesion model control group (PL), the kanglaite control group (KLT), the tamoxifen control group (TAM), the low dose group of ZTO (ZTO-L), the middle dose group of ZTO (ZTO M) and the high dose group of ZTO (ZTO-H). Animal models of precancerous lesion of rats mammary were induced bye DMBA and were treated for 4 weeks. All the animals were killed from 8th to 14th week once every two weeks. Examination of histopathology was performed and the expression of VEGF mRNA was measured by in situ hybridization. RESULTS: The positive rate and strength of expression of VEGF mRNA trended to increase in every groups. In atypycal ductal hyperplasion, the positive rate of expression of VEGF mRNA respectively were (36. 27±5. 36)% (ZTO-L), (30.83±4.12)%(ZTO-M),(26.14±3.17)%(ZTO-H) ,(45.49±2.86)% (KLT), (47.43±6.32)% (TAM) ,(78.65±6.28)% (PL). The strength of VEGF mRNA of intervention group was much lower than made model control groups (P〈0.05),and the strength of VEGF mRNA of ZTO was much lower than KLT and TAM (P〈0.05). In the case of ADH which the expression of VEGF mRNA was positive,the strength of VEGF mRNA of ZTO-H was much lower than ZTO-L (P〈0.05). CONCLUSION: Zedoray Turmeric Oil can decrease the expression of VEGF mRNA in precancerous lesion of rats mammary induced by DMBA, which could be the effective mechanism to inhibits angiogenesis and prevent the breast cancer.
