摘  要:目的探讨ACK1在肺鳞状细胞癌组织中的表达及其临床意义。方法分别采用RT-PCR、免疫组化法检测45名患者肺鳞状细胞癌及癌旁正常黏膜组织标本中ACK1基因mRNA的转录水平和ACK1蛋白的表达水平,并分析ACK1高表达与临床病理特征之间的相关性。结果 60%的肺鳞状细胞癌组织中ACK1基因mRNA转录水平升高;64.4%的肺鳞状细胞癌细胞浆中ACK1蛋白高表达;ACK1高表达与肺鳞状细胞癌的淋巴结转移、肿瘤分期相关(P<0.05);ACK1高表达与肺鳞状细胞癌的不良预后相关,但不是一个独立的影响预后的因素。结论 ACK1可作为一种新型分子标记物,预测原发性肺鳞状细胞癌患者的肿瘤侵袭和淋巴结转移,判断患者预后转归,为肿瘤的个体化治疗提供了实验依据。Objective To investigate the expression of ACK1 in lung squamous cell carcinoma tissue as well as its clinical significance. Methods Transcription level and ACK1 mRNA and expression level of ACK1 protein in lung squamous cell carcinoma tissues and normal mucosal tissues adjacent to the carcinoma of 45 patients were determined by RT-PCR and immunohistochemical assay, based on which the relationship between high expression of ACK1 and clinical pathologic characteristics of lung squamous cell carcinoma was analyzed. Results The transcription levels of ACK1 mRNA increased in 60% of lung squamous cell carcinoma tissue samples. However, ACK1 protein was highly expressed in 64. 4% of cytoplasma of lung squamous cell carcinoma cells. The high expression of ACK1 was associated with the lymph nodes metastasis and stage of lung sqnamous cell carcinoma (P 〈 0. 05 ). Though the high expression was associated with poor prognosis of lung squamous cell carcinoma, it was not an independent factor affecting the prognosis. Conclusion ACK1 might be used as a novel biomarker for prediction of tumor invasion and lymph node metastasis and judgment of prognosis in patients with primary lung squamous cell carcinoma, which provided an experimental basis for individual therapy of tumors.
