摘 要:目的:探讨p16基因和miR-137基因在口腔扁平苔藓(OLP)癌变中的作用。方法:利用甲基化特异性的PCR技术分别检测10例OLP、10例口腔鳞癌(OSCC)新鲜组织样本中p16基因和miR-137基因启动子的甲基化状态,结合临床资料进行分析。结果:在OLP中p16基因和miR-137基因的甲基化率为20%(2/10)和50%(5/10);OSCC中p16基因和miR-137基因甲基化率分别为50%(5/10)和60%(6/10)。两种基因甲基化率无明显差异(P>0.05),由于样本偏少,两种样本中基因甲基化水平并无统计学差异(P>0.05)。p16和miR-137基因甲基化水平与患者年龄、性别以及肿瘤分期分别相关。结论:OLP的上皮组织中存在着p16和miR-137基因的异常甲基化,男性、高龄较易出现p16和miR-137基因的甲基化。Objective:To investigate p16 gene and miR-137 gene methylation in lichen planus mucosa and oral squamous cell carcinoma. Method: 10 patients of oral lichen planus and 10 patients with OSCC were studied on p16 and miR-137 methylation. Result:Rates of p16 and miR-137 methylation were 20 % and 50% for patients of oral lichen planus, 50 % and 60 % for patients of OSCC. There were no statistically significant differences of p16 gene or miR-137 gene methylation rates between two groups. Rates of p16 gene and miR-137 gene methylation were associated with patients' age, gender and stage of the tumor respectively. Conclusion: There were aberrant methylation of p16 gene and miR-137 gene in oral lichen planus tissues. Male, aged patients showed higher methylation rates of p16 gene and miR-137 gene. More samples are needed for further comparasion and indication analysis.
