摘 要:目的:探讨IFN-α和IFN-γ对人肺腺癌细胞株A549 B7-H4 mRNA和蛋白表达的影响。方法:用不同浓度(100、500、1 000、2 000 U.ml-1)IFN-α和IFN-γ分别处理A549细胞,未经药物处理细胞作对照(对照组)。用MTT法观察细胞增殖活性的差异,RT-PCR观察B7-H4 mRNA表达水平的变化,蛋白质印迹法观察B7-H4蛋白表达水平的变化。结果:IFN-α和IFN-γ对人肺腺癌细胞株A549增殖有抑制作用,其抑制效应具有时间和剂量依赖性(P<0.05)。随着IFN-α浓度的增加,B7-H4分子mRNA和蛋白表达增强(P<0.05),而IFN-γ对B7-H4分子mRNA和蛋白表达影响不明显(P>0.05)。结论:IFN-α和IFN-γ均可抑制人肺腺癌A549细胞株的增殖;IFN-α可增强B7-H4mRNA和蛋白水平的表达,而IFN-γ对B7-H4分子mRNA和蛋白表达影响不明显。Objective: To explore the effects of IFN-α and IFN- 3J on B7-H4 expression in human lung adenocarcinoma cell line A549. Methods: Human lung adenocarcinoma A549 ceils were treated with a concentration of 100,500, 1 000, 2 000 Uml-1 IFN-α and IFN-γ,respectively. The inhibitive effect of IFN-α and IFN-γ on proliferation of A549 cells was detected by MTT assay. BT-H4 mRNA expression, by retro-transcriptase polymerase chain reaction (RT-PCR) ; BT-H4 protein expression, by Western blotting assay. Results: TFN-α and IFN-γ inhibited the proliferation of human lung adenocarcinoma cell line A549 in a dose-, time-dependent manner (P 〈 0.05 ). With the increasing in IFN-α concentration, B7-H4 molecule mRNA and protein expression increased (P 〈 0.05 ). But there was no change of B7-H4 molecule mRNA and protein expression with the increasing in IFN-y concentration. Conclusion: IFN-α and IFN-γ may inhibit the growth of human lung adenocarcinoma cell line A549 , and IFN-α may enhance BT-H4 mRNA and protein expression. IFN-T has no effect of B7-H4 mRNA and protein expression in A549.
