摘 要:目的建立梅花入骨丹多糖中蛋白含量的测定方法。方法用水提醇沉法提取梅花入骨丹多糖,考马斯亮蓝法测定蛋白含量,比较Sevage法、木瓜蛋白酶-Sevge法、三氯乙酸-Sevage法、三氯乙酸-正丁醇法脱蛋白的效果。结果 Sevage法、木瓜蛋白酶-Sevage法、三氯乙酸-Se-vage法、三氯乙酸-正丁醇法蛋白去除率分别为46.21%、71.43%、81.12%、88.11%,多糖损失率分别为33.73%、18.51%、43.40%、28.15%。结论木瓜蛋白酶-Sevage法脱蛋白较好。Objective To determine the protein content in polysaccharides from Bauhinia Champi- oni (Benth.) Benth.. Methods The water extraction and alcohol precipitation method was adopted to obtain polysaccharide, Coomassie brilliant blue method was used to detect the protein content. The Sevage method, combined methods of papain and Sevage, combined methods of trichloroacetic acid and Sevage and combined methods of trichloroacetic acid and n-butanol were adopted to remove the protein respectively, and the effects of different methods were compared. Results The percentage of the removed protein for Sevage method, combined methods of papain and Sevage, combined methods of trichloroacetic acid and Sevage and combined methods of trichloroacetic acid and n-butanol were 46.21%, 71.43%, 81.12% and 88.11% respectively, and the loss rate of polysaccharides was 33.73%, 18.51%, 43.40% and 28.15% respectively. Conclusion The combined methods of papain and Sevage was the best for removing protein.
