摘  要:目的:探讨莱菔硫烷(sulforaphane,SFN)对膀胱癌细胞侵袭性及上皮-间质转化(epithelial-mesenchymal transition,EMT)影响和作用机制。方法:常规培养膀胱癌细胞株T24后,采用Transwell侵袭小室法观察SFN对T24细胞体外侵袭能力的影响;采用Western blot法检测SFN作用T24细胞后EMT标志蛋白的表达变化。结果:10μmol/L SFN作用24h后的T24细胞穿透能力显著下降,明显诱导EMT标志蛋白E-cadherin的表达增高并下调vimentin的表达(P<0.05)。结论:SFN能降低膀胱癌细胞的侵袭能力,其作用机制与抑制膀胱癌细胞EMT过程有关。Objective: To investigate the effects of sulforaphane on the invasion and Epithelial-to-Mesenchymal Transition(EMT) in human bladder cancer cell and its mechanism in vitro.Methods: The migration and invasion of sulforaphane on T24 cells were tested by transwell assay.Western blot analysis was used to investigate the protein expression of E-cadherin and vimentin.Results: Sulforaphane(10μmol/L) could dramatically enhance the expression of E-cadherin,and meanwhile reduce the expression of vimentin in T24 cells(P〈0.05).Conclusion: These results suggest that sulforaphane can obviously suppress the invasion of T24 cell.Its mechanism is associated with the Epithelia-to-Mesenchymal Transition.
