摘  要:目的 探讨榄香烯对人胃癌SGC-7901 细胞株增殖以及对细胞外信号调节激酶(ERK1/2)信号通路的影响,并探讨其可能的作用机制.方法 用不同浓度榄香烯(0.01、0.02、0.04mg/ml)处理胃癌SGC-7901细胞,然后用CCK -8法观察细胞增殖抑制作用,光镜和透射电镜观察细胞形态学改变,Western-blot 法检测磷酸化ERK1/2 的表达水平.结果 榄香烯可抑制胃癌SGC-7901细胞的增殖,呈浓度和时间依赖性;光镜和透射电镜下可以观察到典型的细胞凋亡形态学改变;榄香烯作用48h后胃癌SGC-7901细胞磷酸化ERK1/2表达水平受到明显抑制,并呈浓度依赖性,各浓度组与对照组比较,差异均有统计学意义(均P<0.01).结论 榄香烯可以抑制胃癌SGC-7901 细胞的增殖,诱发其凋亡,作用机制可能为抑制了细胞的ERK1/2 信号通路.Objective TO investigate the effect of elemene on the proliferation of human gastric cancer cell line SGC-7901 and the extracellular signal regulated kinasel/2 (ERK1/2) pathway. Methods SGC-7901 cells were treated with various con- centrations of elemene (0.01, 0.02, 0,04mg/ml). Cell proliferation was assayed by CCK-8 method. The morphological changes were observed with light microscopy and transmission electron microscopy. The contents of phosphorylated ERK1/2 were de- tected by Western-blot. Results Elemene had a potent inhibitory effect on the growth in SGC-7901 cells in a dose-dependent and time-dependent manner. The representative morphological changes of apoptosis were seen under the light microscope and transmission electron microscope. Western blot analysis showed that elemene significantly down-regulated the expression of phosphorylatecl ERK1/2 in SGC-7901 cells (P〈0.01). Conclusion Elemene can inhibit the proliferation of gastric cancer SGC-7901 cells and induce apoptosis, which is possibly related to ERK1/2 signal pathway.
