摘 要:目的探讨人胰腺癌组织中血管内皮生长因子C(VEGF-C)及血管内皮生长因子D(VEGF-D)的表达与血管淋巴管生成的关系。方法选用63例根治性手术切除的胰腺癌组织及13例正常胰腺组织,用免疫组织化学(Envi-sion法)检测VEGF-C及VEGF-D在胰腺癌中的表达,并免疫组织化学染色CD31显示计数癌周微血管密度(MVD)和微淋巴管密度(MLVD),进行统计学分析比较胰腺癌中VEGF-C、VEGF-D表达和癌周MVD/MLVD的关系。结果 VEGF-C及VEGF-D阳性表达产物主要见于癌细胞胞浆,胰腺癌周围组织中MVD/MLVD明显升高;胰腺癌中VEGF-C、VEGF-D表达与癌周MVD/MLVD升高呈正相关(P<0.05)。结论胰腺癌中VEGF-C与VEGF-D的表达增高,均具有促进胰腺癌的血管淋巴管增生、促进癌细胞转移的能力。选择性阻断VEGF-C或VEGF-D的促血管生成作用,对控制胰腺癌的淋巴道转移可能是一个有效的选择。Objective To determine vascular endothelial growth factor C(VEGF-C) and D expression in pancreatic carcinomas and the relationship of the expression of VEGF-C and VEGF-D with lymph angiogenesis.Methods The expression of VEGF-C and VEGF-D were detected by immunohistochemical method in 63 cases of radical resected specimen of pancreatic cancer.The microvessel density(MVD) and microlymphatic vessel density(MLVD) of tumor-surrounding tissue were detected by anti-CD31 immunostaining.The normal pancreatic tissue(13 cases) served as the control group.Statistical analysis was used to determine therelationship between the expression of VEGF-C and VEGF-D and MVD/MLVD.Results The expression of VEGF-C and VEGF-D were observed in the cytoplasm of carcinoma cell.The overexpression of VEGF-C and D in pancreatic carcinomas were correlated significantly with high MVD/MLVD of tumor-surrounding tissue(P 0.05).Conclusion The overexpression of VEGF-C and D may be contribute to lymph angiogenesis and cancerometastasis.Inhibiting the effect of VEGF-C or VEGF-D maybe is an effective option to control lymphatic metastasis of pancreatic carcinomas.
