摘 要:目的:研究MGb1-Ag在人体各系统和器官的正常组织和肿瘤中的表达和分布。观察其在胃癌中表达的临床意义。方法:(1)免疫组织化学检测MGb1-Ag在人体各正常组织和常见肿瘤、正常胃粘膜、慢性胃炎、肠上皮化生、不典型增生、胃癌及淋巴结转移灶中的表达和分布,观察胃癌特异性抗原MGb1-Ag在全身各组织器官中的分布,统计分析其在胃癌中表达与临床病理参数之间的关系;(2)激光共聚焦检测MGb1-Ag在胃癌细胞中的定位。结果:MGb1-Ag仅在少数正常组织中有表达,在常见肿瘤中,主要在胃癌、结肠癌、食管癌、胰腺癌、前列腺癌和部分肝癌、肺癌中阳性表达,而在其它肿瘤中表达较少或几乎不表达。MGb1-Ag在胃癌中的表达与胃癌的分化、分期和淋巴结转移密切相关。MGb1-Ag定位于胃癌细胞系SGC-7901的细胞膜上。结论:MGb1-Ag是胃肠道特异性抗原,在胃癌中的表达与胃癌的分化和转移相关,是胃癌新的候选分子标志物。Objective: To Investigate expression and distribution of MGb1-Ag in various normal tissues and organs and malignant tumors.To investigate the clinical value of the MGb1-Ag expression in gastric carcinomas.Methods: ①Immunohistochemical analysis was used to examine the expression of MGb1-Ag in multiple normal or malignant tissues,normal stomach mucosa,chronic gastritis,metaplasia,dysplasia,gastric carcinoma and lymph node metastases.Then the distribution of MGb1-Ag in multiple normal or malignant tissues was observed,and the relationship between the expression of MGb1-Ag in gastric cancer and clinical pathology parameters was statistically analyzed.②The subcellular localization of MGb1-Ag in gastric cancer cells was detected by confocal laser-scanning mi-croscopy.Results: ① MGb1-Ag expressed only in few normal tissues,but positively expressed in several common tumors,such as gastric cancer,colonic cancer,esophageal carcinoma,pancreatic cancer,prostate cancer and some hepatic carcinoma,lung cancer.And MGb1-Ag was negative in all other detected tumors.In addition,the expression of MGb1-Ag correlated with differentiation,stages and lymph node metastasis of gastric carcinoma.② The subcellular localization of MGb1-Ag in gastric cancer cell SGC7901 was mainly in cytomembrane.Conclusion: The expression of gastrointestinal-specific antigen MGb1-Ag correlated with differentiation and metastasis of gastric cancer,so MGb1-Ag could be used as a novel bio-marker candidate.
