摘 要:目的:探讨新基因激活转录因子5(Activating Transcription Factor 5,ATF5)、B细胞淋巴瘤/白血病-2(B-cell lym-phoma/Leukemia-2,Bcl-2)在结直肠癌组织中的表达,探讨其意义及相互关联。方法:选择36例结直肠腺癌组织及自身癌旁正常组织,采用RT-PCR检测两组ATF5、Bcl-2 mRNA的表达情况,并分析其表达与肿瘤分化程度的相关性。结果:ATF5、Bcl-2在结直肠腺癌组织中的表达均高于癌旁组织(P<0.01);ATF5、Bcl-2 mRNA表达高低与结直肠腺癌细胞分化程度相关,ATF5 mRNA在低分化组表达高于高分化组(P<0.05),Bcl-2 mRNA在高分化组表达高于低分化组(P<0.05);ATF5、Bcl-2 mRNA在结直肠腺癌组织内表达呈负相关。结论:ATF5在结直肠癌的发生、发展中具有一定作用,其作用机制可能与调节其下游基因Bcl-2的表达有关。Objective: To investigate the expression of Activating Transcription Factor 5(ATF5) mRNA and B-cell lym-phoma/Leukemia-2(Bcl-2) mRNA in colorectal adenocarcinoma.Methods: RT-PCR was used to examine the expression level of ATF5 and Bcl-2 mRNA and in 36 colorectal adenocarcinoma tissues and adjacent tissues.Then the relationship between ATF5 and Bcl-2 was analyzed.Results: The mRNA expression level of ATF5 and Bcl-2 in colorectal adenocarcinoma tissues were higher than the adjacent tissues(P〈0.01),and in the adenocarcinoma tissues,the mRNA levels were related to the differentiation of the colorectal tumors.More ATF5 mRNA was detected in poorly differentiated adenocarcinoma tissues than well/moderately differentiated adenocarcinoma tissues while the Bcl-2 with a completely opposite result.A negative correlation between ATF5 and Bcl-2 was found in the colorectal adenocar-cinoma tissues.Conclusion: The abnormal expression of ATF5 may play an important role in the development and progression of col-orectal adenocarcinoma.ATF5 may play it’s role as a inhibitor of apoptosis by regulating the expression of Bcl-2.
