摘 要:目的探讨扭曲肉芝甲酯抗结肠癌LoVo细胞增殖活性及其机制。方法将LoVo细胞随机分为对照组和处理组(10、30、50umol/L3个亚组)。对扭曲肉芝甲酯处理后LoVo细胞凋亡率、细胞周期、凋亡蛋白表达的改变进行定量分析。结果经10、30、50umol/L扭曲肉芝甲酯处理24h后,LoVo细胞凋亡率分别为(2.32±0.93)%、(11.03±0.14)%和(16.29±1.98)%,对照组凋亡率为(2.09±0.97)%,而50umol/L扭曲肉芝甲酯处理0、6、12、24h后凋亡率分别为(2.67±0.97)%、(5.08±1.03)%、(5.04±0.83)%和(13.50±1.16)%(P〈0.05)。荧光显微镜下细胞呈现典型的凋亡性改变;扭曲肉芝甲酯在0、10、50及100umoL/L时将LoVo细胞抑制在G2/M期(9.53±3.97)%、(10.26±1.89)%、(11.36±2.56)%、(15.39±1.56)%(P〈0.05),该期的比例逐渐增高;Westernblot法结果显示扭曲肉芝甲酯处理后结肠癌kVo细胞的半胱氨酰天冬氨酸特异性蛋白酶(Caspase)-8、Caspase-3表达升高。结论扭曲肉芝甲酯以浓度依赖性和时间依赖性的方式抑制结肠癌LoVo细胞增殖,抑制细胞周期G2/M期,通过Caspase-8/Caspase-3来诱导细胞凋亡。Objective To investigate methyl sartortuoate anti LoVo cells proliferative activity and the mechanism involved. Methods LoVo cells were randomly divided into control and treatment groups (10, 30, 50 umol/L), methyl sartortuoate process LoVo ceils apoptosis rate, cell cycle, apoptosis, protein expression changes, quantitative analysis. Results The methyl sartortuoate in a certain concentration range, concentration-dependent and time-dependent manner to inhibit the growth of LoVo cells (P 〈 0. 05). After methyl sartortuoate 0, 10, 30 and 50 p, mol/L for 24 h, the LoVo cell apoptosis rate was ( 2. 09 ± 0. 97 ) %, ( 2. 32 ± 0. 93 ) %, ( 11.03 ± 0. 14 ) % and ( 16. 29 ±1.98 ) %, 50 umol/L methyl handle processing 0, 6, 12, 24 h. The apoptosis rates were (2. 67 ±0. 97)%, (5.08 ± 1.03)%, (5.04 ±0. 83)% and ( 13. 50 ± 1.16) % ( P 〈 0. 05 ) respectively. The ceils showed typical apoptotic changes in the fluorescence microscope; LoVo ceils 0, 10, 50 and 100 umol/L, inhibition at the G2/M phase (9.53 ± 3.97)%, (10.26±1.89)%, (11.36±2.56)%, (15.39 ±1.56)% (P〈0.05), the proportion of gradually increased. Western blotting results showed that methyl sartortuoate processing of LoVo cells caspase-8 and caspase-3 expression was elevated B lymphocytes/leukemia-2 ( bcl-2 ) expression decreased. Conclusion The methyl sartortuoate can inhibit the proliferation of colon cancer LoVo cell in concentration-dependent and time-dependent manner, and induce cell apoptosis through caspase-8/caspase-3.
