吉西他滨联合奥沙利铂对人胆管癌裸鼠移植瘤模型的药效实验研究

摘　　要：目的观察吉西他滨联合奥沙利铂对荷人胆管癌裸鼠移植瘤的治疗效果。方法培养人胆管癌裸鼠移植瘤模型,将49只荷人胆管癌裸鼠分成7组,每组7只,分别为空白对照组,奥沙利铂(A)组,吉西他滨(B)组,A+B组,2A+B组,A+2B组和2A+2B组,于分组当天腹腔注射奥沙利铂,第2天注射吉西他滨。给药剂量:奥沙利铂1.125mg/kg,吉西他滨50mg/kg,生理盐水0.3mL/只。第24天处死裸鼠后测量肿瘤标志物,进行移植瘤病理及流式细胞仪分析。治疗期间每周2次测量肿瘤直径,计算相对肿瘤增殖率。结果荷人胆管癌裸鼠移植瘤成瘤率100%。A、B组第24天相对肿瘤增殖率分别为39.78%和45.73%;2A+2B组相对肿瘤增殖率最低,为32.05%。治疗组与空白对照组相比,肿瘤标志物、病理分析及MTT分析均有显著差异。结论吉西他滨、奥沙利铂对荷人胆管癌裸鼠移植瘤有抑制作用,两药联合有助于提高疗效。Objective To observe the effect of gemcitabine combined oxaliplatin on nude mice bearing hu- man bile duct carcinoma. Methods To raise the human bile duct carcinoma nude model. 49 nude mice were divided into 7 groups as control group, oxaliplatin（A） group, gemcitabine（B） group, A ＋ B group, 2A ＋ B group, A ＋2B group, 2A ＋2B group. Each group had 7 mice. On the day of classification, oxaliplatin was injected via abdominal cavity, and the second day, gemcitabine was injected. The doses of drugs administrated were as follows： A： l. 125mg/kg,B： 50nlg/kg, 0.3 ml of physiological saline solution was used for the control mouse. At the end of the experiment, the mice were killed for determination of tumor markersand microscopic exanfination. During the experiment, tumor sizes were measured twice a week and relative tumor proliferation rates were calculated. Results Nude mice bearing human bile duct carcinoma were all raised. Relative tumor proliferation rates of A group and B group were 39.78% , 45.73%. Relative tumor proliferation rate of 2A ＋ 2B group was 32.05% , it was minimum. There was significant deviation between treated group and control group in the tumor markers and microscopic examination. Conclusions Gemcitabine and oxaliplatin exhibit effect on nude mice bearing human bile duct carcinoma, the combination of gemcitabine and oxaliplatin can conduce to sublime therapeutic effect.
