摘  要:目的建立抗甲型流感病毒作用新的鸡胚模型。方法设计尿囊腔中接种流感病毒(甲型流感病毒鼠肺适应的FM1株及流感病毒A3),鸡胚卵白注射利巴韦林,35℃培养72 h后,观察尿囊液流感病毒血凝素滴度。结果在新的模型下,利巴韦林100、50、25 mg/kg剂量组对流感病毒FM1及流感病毒A3在鸡胚体内有明显的抑制作用,具有统计学意义(P<0.05)。感染前2 h及感染后0 h、2 h、4 h注射利巴韦林,对流感病毒FM1及流感病毒A3在鸡胚体内有明显的抑制作用,具有统计学意义(P<0.05)。结论在新的鸡胚模型中,利巴韦林可明显地抑制流感病毒的复制。Objective To develop an embryonated chicken egg model for in vivo evaluation of anti-influenza activity.Methods The influenza virus A was injected into the amniotic cavity and d a single dose of ribavirin was injecte into the albumen at the sharp pole of the embryonated chicken egg instead of into the amniotic cavity.After incubated in 35℃ for 72 h,the virus titer was determined by hemagglutination test.Results In the new chicken embryo model,ribavirin was delivered from the albumen to allantoic cavity successfully and no toxic effects at therapeutic concentrations of the ribaririn were found: In the new chicken embryo model,ribavirin treatment at the concentrations of 25 mg/kg to 100 mg/kg dramatically decreased hemagglutination titer of both IFVFM1 and IFVA3: In the new chicken embryo model,the addition of ribavirin at 4 hour post-infection or earlier led to marked suppression.However,there appeared to be no significant differences in 6h and 8h group compared with untreated group: In the new chicken embryo model,the ribavirin′s antiviral effect can be tested under the different challenge dose of IFVFM1 and IFVA3.Conclusion The model could be an alternative model for in vivo evaluation of activity against Influenza virus A.
