摘  要:目的:采用腺相关病毒(Adeno-associated virus,AAV)-9为载体,比较3种转染方法对ZsGreen基因在大鼠心肌组织表达的影响。方法:30只雄性SD大鼠随机分为尾静脉(Tail vein,TV)注射组、心肌(Intramyocardium,IM)注射组、冠脉(Intracoro-nary,IC)灌注组,采用相应方法在体转染携带报告基因ZsGreen的腺相关病毒-9型(Adeno-associated virus 9-ZsGreen,AAV9-ZsGreen)。2周后荧光显微镜下观察绿色荧光在大鼠IM组织的表达情况。结果:TV组IM有少量绿色荧光分布;IM组绿色荧光表达主要局限在IM注射位点,注射点周边有少量分布;IC组大量绿色荧光在IM广泛均匀表达。IM及IC组荧光强度均显著高于TV组(P<0.01),IC组荧光强度高于IM组(P<0.01)。结论:IC灌注法可使AAV介导的外源基因在IM组织高效、广泛、均匀表达,优于TV注射法及IM注射法。Objective:To compare the effect of three transfection methods on ZsGreen gene expression in rats myocardium mediated by adeno-associated virus 9(AAV9).Methods:Totally 30 male SD rats were divided into tail vein(TV) injection group,intramyocardium(IM) injection group and intracoronary(IC) infusion group.AAV9 carrying report gene ZsGreen were transferred into rats'myocardium by methods mentioned above and green fluorescence expression in myocardium was observed by fluorescence microscopy two weeks later.Results:A little green fluorescence expression could be observed in myocardium of TV injection group;in IM injection group,most green fluorescence expressions were localized at the injection sites and a small amount aroud the injection sites;in IC infusion group,there was large green fluorescence in myocardium of broad and uniform distribution.The fluorescence intensity was increased significantly in IM injection and IC infusion group compared with that in TV injection group(P〈0.01).The fluorescence intensity in IC infusion group was higher than that in IM injection group(P〈0.01).Conclusion:IC infusion can make AAV-mediated ZsGreen gene express in an efficient,extensive and well-distributed way in myocardium tissues,superior to TV injection and IM injection.
