摘 要:采用高效液相色谱(HPLC)分析漳州水仙‘金盏银台’花瓣和副冠中主要类胡萝卜素成分。结果显示,黄色副冠的主要色素成分是叶黄素,含量为3.73×104μg/g,β-胡萝卜素的含量很低,仅2.7μg/g;白色花瓣中各种类胡萝卜素的含量都极低。同时,采用RT-PCR方法分析类胡萝卜素合成途径结构基因PSY、PDS、ZDS、LYCE、LYCB、BCH在‘金盏银台’白色花瓣、黄色副冠,洋水仙‘粉红魅力’白色花瓣、红色花副冠,洋水仙‘嘹亮’黄色花瓣、橙黄色副冠中的表达情况。结果表明,花瓣与同一品种副冠间结构基因的表达相似,‘粉红魅力’红色副冠中PDS表达增强,而ZDS、LYCE、BCH的表达减弱。从结果可以看出,漳州水仙花中,类胡萝卜素合成主要是叶黄素代谢分支;推测漳州水仙花瓣呈现白色与质体的结构和数量有关;‘粉红魅力’红色副冠中由于几个结构基因的共同作用,类胡萝卜素合成转向β-胡萝卜素分支并累积β-胡萝卜素。To evaluate the carotenoid components, the extracts from perianths and coronas of Zhangzhou narcissus (Narcissus tazetta vat. chinensis) 'Jinzhanyintai' were analyzed separately by HPLC. The results showed that the most abundant carotenoid pigment in yellow corona was lutein, 3.73×104μg/g in content and the content of β- carotene was very low, only 0.00027×104μg/g. The content of each carotenoid component was little in white perianths of Zhangzhou narcissus. The expression of carotenoid biosynthetic genes in Zhangzhou narcissus, Narcissus pseudonarcissus 'Pink charm' and'Fortissimo' was also investigated, including phytoene synthase (PSY), phytoene desaturase (PDS), X-carotene desaturase (ZDS), lycopene β-cyelase (LYCB), lycopene e-eyclase (LYCE) and β-carotene hydroxylase (BCH). The similar expression between perianths and coronas of the same variety and changes in the expression of PDS, ZDS, LYCE and BCH between 'Pink charm' and other two varieties were observed. These resuhs suggest that lutein biosynthetic pathway is the main one in the corona of Zhangzhou narcissus, the lutein biosynthetic pathway is replaced by β-carotene pathway and β-carotene accumulated in the red corona of 'Pink charm' with the co-action of PDS, ZDS, LYCE and BCH genes. One possible explanation for the white of perianth in Zhangzhou narcissus is the difference in the structure and number of plastid. The results of this study will yield insights into the carotenoid biosynthesis in Chinese narcissus.
