摘  要:目的探讨激光氧液(LOF)对戊四氮(PTZ)诱导癫痫反复发作大鼠海马谷氨酸(Glu)和γ-氨基丁酸(GABA)含量变化的影响。方法 SD大鼠200只随机分为模型糖水组(PTZ+GS组)、模型激光氧液组(PTZ+LOF组)、盐水糖水组(NS+GS组)、盐水激光氧液组(NS+LOF组)和空白组(Sham组)五个亚组,选择处理后当天(0d)、3d、7d、10d和14d时间点进行观察,应用高效液相方法检测海马组织Glu和GABA含量。结果 PTZ+LOF组大鼠经过激光氧液治疗后7~14d惊厥发生率显著下降,在第14天时比治疗前下降50%,同PTZ+GS组比较在治疗7~14d时两者表达差异均有统计学意义(P<0.05)。PTZ+GS组和PTZ+LOF组发作后海马组织Glu的含量较NS+GS组、NS+LOF组和Sham组有显著增高(P<0.05),PTZ+LOF组各时间点Glu含量较PTZ+GS组下降(P<0.05),以3d下降趋势明显;GABA含量在PTZ+GS组和PTZ+LOF组各时间点浓度也大于NS+GS组、NS+LOF组和Sham组(P<0.05),点燃大鼠经LOF治疗后海马中GABA含量较PTZ+GS组明显升高,其中在7d、10d、14d三个时间点上较PTZ+GS组差异有统计学意义(P<0.05)。结论 LOF能够降低大鼠癫痫发作后异常增加的Glu含量,升高GABA含量。Objective To explore the effect of laser oxygenate fluid on the changes of contents of glutamate ( Glu ) and gamma-aminobutiric acid ( GABA ) in hippocampus formation in Pentylenetetrazol ( PTZ ) induced epilepsy rats. Methods SD rats ( n = 200) were randomly divided into PTZ + GS group, PTZ + LOF group, NS + GS group, NS + LOF group and normal group. The day after the selection process (0 day) ,3 days,7 days,10 days and 14 days time point observation, the application of high performance liquid methods detection hippocampus formation organization Glu and GABA content. Results Change of content of Glu and GABA in homogenized brain tissue of the rats after epilepsy :PTZ + LOF rats ,liquid oxygen treatment 'after laser 7-14 d convulsions rate decreased significantly ,the 14 d in before treatment than dropped to 50% ,compared with PTZ + GS group in the treatment of 7-14 d both expression differences are statistically significant ( P 〈 0.05 ). Content of Glu was higher 'after epilepsy in PTZ + GS group, PTZ + LOF group than in NS + GS group, NS + LOF group and normal group ( P 〈 0. 05 ), the content of Glu in PTZ + LOF group was decreased than PTZ + GS group ( P 〈 0. 05 ). To 3 days down trend evident ; GABA content in PTZ + GS group and PTZ + LOF group of each time point the concentration than NS + GS group, NS + LOF group and Sham group (P 〈 O. 05 ), lit by LOF rats 'after treatment in the hippocampus GABA content is PTZ + GS group increased significantly, which in a 7 d, 10 d, 14 d three time points on relatively PTZ + GS group (P 〈 0.05 ). Conclusions LOF can reduce the content of Glu which is abnormally increased and rise GABA content in epileptic seizure rats.
