摘  要:目的探讨Stat3及Bcl-2、Bax在食管上皮内瘤癌变过程中的可能作用。方法采用免疫组化方法检测食管上皮内瘤变(150例)和食管鳞癌(100例)组织中Stat3、Bcl-2、Bax在蛋白水平的表达情况。结果 Stat3蛋白在正常食管黏膜、食管上皮内瘤变、食管癌病例中的阳性表达率分别为6.67%、60.67%、94.00%,食管上皮内瘤变和食管癌病例中的Stat3蛋白的表达明显高于正常食管黏膜组(P<0.05),且食管癌中Stat3蛋白的表达高于食管上皮内瘤变组(P<0.05);Stat3蛋白和Bcl-2蛋白的表达均随食管内瘤变级别的增高及食管癌的出现而增高(P<0.05),但Bax蛋白的阳性表达率随食管内瘤变级别的增高及食管癌的出现呈降低趋势。Stat3与Bcl-2的表达呈正相关(P<0.05),但Stat3与Bax的表达不存在相关性(P>0.05)。结论 Stat3及Bcl-2、Bax基因在食管鳞癌的发生、发展中均起到一定作用,Stat3和Bcl-2可能在食管癌变发生过程中起重要的协同作用。Objective To explore the possible roles of Stat3 and Bcl-2,Bax in carcinogenesis of esophageal intraepithelial neopla sia. Methods The expression of Stat3 ,Bcl-2 and Bax at protein level was studied in 150 cases of esophageal intraepithelial neoplasia and 100 cases of esophageal squamous cell carcinoma by immunohistochemical staining. Results The positive expression rate of Stat3 was 6.67 %, 60.67 %, 94.00% respectively in normal esophageal mucosa, esophageal intraepithelial neoplasia and esophageal carcinoma. The positive expression of Stat3 in the cases of esophageal intraepithelial neoplasia or esophageal carcinoma was all significantly higher than that in normal esophageal mucosa(P〈0. 05) ,and the positive expression of Stat3 was significantly higher in esophageal carcinoma than that in esophageal intraepithelial neoplasia(P〈0.05). The positive expression of Stat3 and Bcl-2 was increased as the degree of esophageal intraepithelial neoplasia increased and esophageal carcinoma occurred(P〈0.05) ;Bax was de scended as the degree of esophageal intraepithelial neoplasia increased and esophageal carcinoma occurred;Stat3 and Bcl 2 were positively related(P〈0.05) ,but Stat3 and Bax were not related(P2〉0.05). Conclusion In the occurrence and development of esopha geal squamous cell carcinoma, Stat3, Bcl-2 and Bax genes play a role, Star3 and Bcl-2 may become synergistic action during the change in esophageal squmous cell carcinoma.
