摘 要:目的:观察层黏连蛋白(laminin,LN)在大鼠磨牙成釉器、牙囊和牙龈组织中的时空表达,并探讨其与上皮细胞形态和极性变化之间的关系。方法:雄性Wistar幼鼠、青春期鼠和成年鼠各4只,取上颌磨牙区牙周-牙体联合组织块,免疫组化PV法显示LN在成釉器、牙囊和牙龈组织中的时空表达,同时观察上皮细胞形态和极性。结果:LN不连续线形表达于成釉器外釉上皮-牙囊间叶组织界面,细胞间存在LN阴性表达间隙;连续线形表达于牙龈上皮和血管内皮基底膜内;散在颗粒形表达于早期牙胚牙囊间充质细胞外基质内和牙龈上皮基底膜下个别牙龈成纤维细胞外基质内。LN表达强度随大鼠齿龄的增加而逐渐下调,成年期基本稳定,上皮细胞形态和极性也发生相应的变化。结论:LN在大鼠不同齿龄成釉器、牙囊和牙龈组织中的表达方式和表达强度不同。LN在组装牙龈上皮基底膜的同时,其表达强度可能受控于牙龈上皮细胞的形态和极性。Objective:To observe the temporal and spatial expression of laminin(LN)in enamel organ,dental sac and gingiva of rat molars,and the shape and polarity of epithelium cells in enamel organ and gingiva.Methods: Twelve male Wister rats(4 young rats,4 adolescent rats and 4adult rats)were killed.The tissue blocks that contained maxillary molars were obtained.The expression of LN was detected by immunohistochemisty of PV.Results: LN was discontinuously and linearly expressed between outer enamel epithelium of enamel organ and mesenchyme of dental sac.Granule-like LN was observed in extracellular matrix around mesenchymes in dental sac of early dentary germ.Continuous and linear LN was shown in basement membrane underneath gingival epithelium and vascular endothelium,occasionally in extracellular matrix around gingival fibroblasts.The expression of LN weakened gradually until adult period when it became stable.At the same time,the shape and polarity of epithelium cells changed correspondingly.Conclusion:At different ages of teeth,LN in enamel organ,dental sac and gingiva expressed differently in shapes and intensity.While LN is assembled into the basement membrane of gingival epithelium,its expression may possibly be influenced by the shape and polarity of gingival epithelium cells.
