摘 要:电压门控质子通道Hv1具有调节细胞内pH值的功能,用RT-PCR、细胞划痕和体外侵袭实验研究Hv1对脑胶质瘤细胞SHG-44迁移和侵袭的影响.RT-PCR结果表明,Hv1在SHG-44中表达;用针对Hv1基因的小RNA干扰片段(SiRNA)下调Hv1基因在SHG-44细胞中的表达明显减弱细胞的迁移和侵袭能力.明胶酶谱实验结果证明,下调Hv1基因在SHG-44细胞中的表达抑制了MMP-2、MMP-9的活性.以上结果表明,Hv1通过调节SHG-44细胞内的pH值来提高SHG-44细胞的迁移和侵袭能力.Voltage-gated proton channel (Hvl) can regulate intracellular pH value. The effects of Hvl gene on the migration and invasion of glioma cell line SHG-44 using RT-PCR, scratch and Matrigel invasion methods were studied. The results showed that Hv1 is expressed in SHG-44 by RT-PCR. The migration and invasion of glioma cells were significantly attenuated when Hvl was knocked down by SiRNA-Hv1. The results of eymographic analysis of matrix metalloproteinase indicated that the enzyme activities of MMP-9 and MMP-2 were attenu- ated when the reduction of Hvl expression in SHG-44 by SiRNA-Hv1. The results showed that Hvl promotes the migration and invasion of SHG-44 through regulating the intracellular pH value.
