摘　　要：目的观察BMP-2基因在静脉移植模型中的动态表达及与内膜增生的关系。方法建立兔自体静脉移植动物模型,分别于术后3、7、14、28d取材。Verhoeff染色观察各时间点内膜厚度及管壁厚度;Western blot检测BMP-2蛋白的变化,免疫组化(SABC)法检测BMP-2、增殖细胞核抗原(PCNA)的表达。结果术后7d内膜明显增厚,14d内膜增厚达到高峰(P<0.01)。BMP-2蛋白在正常对照血管可见表达,术后3d内无表达,术后第七天开始表达,第十四天明显增高,28d达到高峰(P<0.01)。PC-NA的表达于术后7～14d达到高峰。结论 BMP-2基因的表达可能是抑制移植静脉内膜增生的环节之一,可能成为治疗移植血管再狭窄的目的基因。Objective To investigate the expression of BMP- 2 protein and its relations with intimal hyperplasia in vein grafts model. Methods Autogenous vein graft model was established in 24 rabbits by transplanting internal branch of jugular vein to carotid artery. The vein grafts were harvested on 3 days, 7 days, 14 days and 28 days respectively after the operation. Intimal hyperplasia （IH） was observed by His- tomorphological method at different time points. Western blot and immunohistochemistry were used to detect the protein production of BMP- 2. Proliferating cell nuclear antigen （PCNA） also was studied. Results After operation, the intimal thickness increased on 7th day compared with controls （P〈 0. 05）, and reached a peak on 14th day. Protein level of BMP - 2 increased significantly on 7th day, peaked on 28th day after operation. The expression of PCNA reached peak at 7-14days. Conclusion The inhibition of BMP - 2 present in autogenous may become a new target for the gene therapy of intimal hyperplasia and stenosis after vein graft.
