ZFYVE26在肝细胞癌中的表达及其意义

摘　　要：目的:探讨ZFYVE26在肝癌细胞株及肝细胞癌组织中的表达情况,为阐明ZFYVE26在肝癌发生发展中的作用提供依据。方法:应用荧光定量PCR方法检测正常肝脏细胞株L-02和肝癌细胞株MHCC-LM3中ZFYVE26mRNA的表达情况。同时采用荧光定量PCR、Western Blot方法检测ZFYVE26在7例人肝细胞癌及其癌旁组织中的表达情况。结果:与L-02对比,ZFYVE26mRNA在MHCC-LM3中的表达量明显降低,两组比较差异有统计学意义(P<0.05);在肝细胞癌中,ZFYVE26基因及其蛋白的表达量均显著低于在癌旁组织中的表达(P<0.05)。结论:ZFYVE26在肝癌细胞株及肝细胞癌组织中的表达水平均明显降低,提示ZFYVE26的低表达与肝癌的发生发展有关。Objective To study the expression of ZFYVE26 in human hepatoma cell line and hepatoeellular carcinoma （HCC）, which could provide basis for further study on the functions of ZFYVE26 in the occurrence and development of HCC. Methods The mRNA expressions of ZFYVE26 in MHCC-LM3 and in human normal liver L-02 cells were detected by real-time quantitative PCR （qRT-PCR）. The mRNA expression and protein levels of ZFYVE26 were detected by qRT-PCR and western blot in 7 cases of HCC and adjacent non-tumorous tissue （NT）. Results Compared to the L-02, the mRNA expression of ZFYVE26 was significantly decreased in MHCC-LM3 and there was significant in statistics （P 〈 0.05）. The mRNA expression of ZFYVE26 and its protein in HCC was down-regulated compared to those in NT （P 〈 0.05）. Conclusion The expression of ZFYVE26 in HCC and MHCC-LM3 are significantly lower than that in NT and L-02 respectively. These results indicate that the expression level of ZFYVE26 is related to the occurrence and development of HCC.
