摘 要:目的:探讨硫化氢(H2S)对铁超载下大鼠肝星状细胞(HSC-T6)氧化应激损伤的保护作用。方法:将HSC-T6分为4组,空白组、次氮基三乙酸铁(FeNTA)组、硫氢化钠(NaHS)组、格列苯脲(GLBN)组。铁超载法复制HSC-T6氧化应激损伤模型,按实验设计分别给予不同剂量NaHS和GLBN,检测24h和48h后上清液中SOD、GSH、TAOC、MDA、GSSG、.OH含量。结果:与FeNTA组比较,NaHS组SOD、GSH、TAOC随剂量呈不同程度的升高(P<0.05),MDA、GSSH、ROS则呈不同程度的降低(P<0.05);GLBN组SOD、GSH、TAOC随剂量有不同程度的降低(P<0.05),MDA、GSSG、ROS则有不同程度的升高(P<0.01)。结论:H2S对铁超载下大鼠HSC-T6具有抗氧化作用,其机制可能与KATP通道开放,降低钙超载有关。Objective To investigate the protective effect of H2S on iron overload oxidative stress injury in rat HSC-T6. Methods HSC-T6 cells were divided into four groups:normal control group, FeNTA group, NariS groups and GLBN groups. HSC-T6 oxidative stress model was set up by using iron overload method. Different doses of NariS and GLBN was used according to experimental design, levels of SOl), GSH, TAOC, MDA, GSSG and ~ OH were detected at 24 h and 48 h after treatment. Result Compared with FeNTA group, levels of SOD, GSH and TAOC increased (P 〈 0.05), but levels of MI)A, GSSH and ROS decreased in NariS group (P 〈 0.05). However, levels of SOD, GSH, TAOC decreased(P 〈 0.05), but MDA, GSSH and ROS levels increased in GLBN group compared with those in FeNTA group (P 〈 0.01). Conclusion H2S protects rat HSC-T6 cells against iron overload-induced oxidative stress
