摘 要:目的:研究加味归脾汤对人类乳腺癌细胞的抑制效果及原理。方法:不同浓度加味归脾汤作用MDA-MB-435细胞,阳性对照药为Docetaxel,通过MTS、细胞凋亡分析和Caspase-8检测等评价药物的抑制效果。结果:加味归脾汤高浓度组的抑制率在48 h时高于Docetaxel,72 h时与Docetaxel相同。流式细胞仪72 h检测结果显示,加味归脾汤高浓度组和Docetaxel总凋亡程度相近。Caspase-8表达量检测显示,加味归脾汤高浓度组在48 h、72 h时均高于Docetaxel。结论:加味归脾汤高浓度组的抑制效果可以达到或超过Docetaxel,其抑制作用可能是通过Caspase途径起效。Objective:To observe the inhibited effects and principles of Modified Spleen Prescription, against breast cancer cells. Methods: The MDA-MB-435 breast cancer cells were inhibited by Modified Spleen Prescription different concentration to evaluate their inhibited effects respectively and investigata its principles synthetically by the MTS, the apoptosis analysis by flow cytometry and the Caspase-8 tests. The positive control medicine was Docetaxel. Results:The inhibited effect of Modified Spleen Prescription was higher than the Docetaxel at 48h and identical at 72 h. The Flow cytometry showed that the degree of apoptosis was similar between the Modified Spleen Prescription of high concentration and Docetaxel. The Caspase-.8 showed that the Modified Spleen Prescription of high concentration was higher than the Docetaxel at 48 h and 72 h. Conclusion:The inhibited effect of Modified Spleen Prescription of high concentration can meet or exceed the Docetaxel. One of the effective pathway is Caspase.
