摘  要:目的:探讨灵芝对人肝癌干细胞糖代谢及标志物CD133蛋白的影响。方法:培养人肝癌SMMC7721细胞并用灵芝L08处理,用MTT法测定细胞的成活率,用试剂盒法测定己糖激酶活性,用Western Blot法测定CD133蛋白的水平。结果:灵芝L08处理人肝癌SMMC7721细胞后,细胞的成活率及己糖激酶活性都有所降低。同时,肝癌干细胞的CD133蛋白水平也降低。结论:灵芝L08能抑制人肝癌细胞的增殖、细胞中己糖激酶的活性和CD133蛋白水平。Objective: To explore the influence of ganoderma to carbohydrate metabolism and marker CD133 protein in human liver cancer stem cells. Methods: Human liver cancer SMMC7721 cells were cultured and treated with ganoderma L08.The numbers of living cells are detected by MTT method.The activity of hexokinase and the level of CD133 protein were measured by kit of hexokinase and by western blot method,respectively. Results: After treat the cell of SMMC7721 cell lines with ganoderma L08,the number of living cells and the activity of hexokinase were decreased.ganoderma L08 also decreased the level of CD133 protein in human cancer stem cells. Conclusion: Ganoderma L08 could inhibit the cell growth and the activity of hexokinase and the level of CD133 protein in liver cancer stem cells.
